Objective:Eosinophils originate from bone marrow stem cells and differentiate in bone marrow.Eosinophils have the abilitities of anti-parasitic and participation in hypersensitivity.In order to explore the role of eosinophils in tumor immunity.We investigated the antitumor effect on K562 and MCF-7 cells,and investigated the interaction between lymphocyte and eosinophil in vitro.Methods:(1)Guinea pig was established of typeⅠhypersensitivity and EOS was isolateed from peripheral blood and peritoneal fluid.HE stain was used to observe the guinea pig lung and pleen tissues.(2)The level of cytokine(TNF-a,IL-4) in guinea pig blood serum and SELC were measured with Radioimmunoassay.(3)MTT assay was used to exmine the effect of EOS,SELC and LLE on the proliferation of K562 and MCF-7.Results:1.It was higher that the OD of the spleen lymphocyte of guinea pig after treated with peripheral blood and peritoneal fluid EOS(1×103/ml,1×104/ml,1×105/ml)for 48h,,the PI most high score reached 1.92,2.58.As well as the content of TNF-a and IL-4 in blood serum and SELC was obviously enhanced(P<0.05).2.K562 and MCF-7 cells were treated by SELC12,SELC24,SELC48.The multiplication of K562 and MCF-7 cells were obviously inhibited by 1×105/ml SELC48after 48h (P<0.05).3.The guinea pig peripheral blood EOS(1×103/ml,1×104/ml,1×105/ml)cocultured with K562.The multiplication of K562 was not inhibited after 48h(P>0.05).But the multiplication of K562 was obviously inhibited after 24h by peritoneal fluid EOS(1×103/ml,1×104/ml,1×105/ml)(P<0.05).The RI most high score reached 53.33%.The inhibitory action gradually strengthened along with peritoneal fluid EOS number increasing progressively.4.The guinea pig peripheral blood EOS(1×103/ml,1×104/ml,1×105/ml)cocultured with MCF-7.The multiplication of MCF-7 was obviously inhibited by peripheral blood EOS(P<0.01).The RI most high score reached 37.80%;The multiplication of MCF-7 was obviously inhibited by peritoneal fluid EOS(P<0.01).The RI most high score reached 28.98%.5.K562 and MCF-7 cells were treated by LLE.It was appeared the obvious change of K562 cells under the microscope.The cellular form was not neat,the edge was rough,cytoplasm was muddy;It was also appeared the obvious change of MCF-7 cells.The cellular form was rough,the arrangement was loose,appeared falls off.6.K562 and MCF-7 cells were treated by LLE.The multiplication of K562 was obviously inhibited after 48h by LLE(EOS cell count is 1×105/ml)(P<0.05).The RI most high score reached 26.13%.But the multiplication of MCF-7 was obviously inhibited by LLE from 12h,and assumed the dosage time dependence(P<0.05).Conclusion:1.EOS may promote the lymphocyte multiplication activeness,enhance the content of TNF-a and IL-4 in blood serum and SELC.2.The multiplication of K562 may be inhibited by SELC,LLE and peritoneal fluid EOS in vitro.3.The multiplication of MCF-7 may be inhibited by SELC,LLE,peripheral blood and peritoneal fluid EOS in vitro.
