Preventive Effect Of Hepatocyte Growth Factor Gene Mediated By Attenuated Salmonella On The Stress Ulcer In Rats

Objective To explore the preventive effect of hepatocyte growth factor gene mediated by attenuated Salmonella on the stress ulcer model by water immersion and restraint stress in rats.Methods 40 Wistar male rats were randomly divided into 4 groups:normal group,model group,prevention group,control group,10 each group.Before stressing, rats in prevent group and in control group were treated by intragastric administration with 1×10⁹cfu/ml of Typl-HGF and Ty respectively,0.3ml each animal,once another day,6 times together.Same volume of 10%NaHCO₃ was administrated to the model and normal control rats.The stress ulcer model of rats in model group,prevention group and control group were established by water immersion and restraint stress(WRS),after 18 hours,all rats were killed.The concentration of NO,Ang-â…¡,ET-â… ,COR,IL-2,IL-6,and TNF in rat serum and plasm was detected by chromatometry and radioimmunity,respectively.The histological changes and degree of injury of gastric mucosa were observed grossly and under light microscope.UI was evaluated.Results:Severe mucosal bleeding and erodion was observed in rats from model group.But in rats from prevention group,the changes were evidently ameliorated,and were significantly milder than that of in rats from control group.The ulcer index(UI)of rats in the prevention group(16.20±5.69)was significantly lower than that of rats in control group(28.10±3.67),model group(42.20±6.71)(P＜0.01).The level of NO,COR,IL-6 and TNF in serum,ET and Ang-â…¡in plasm in model group were higher than that normal group(p＜0.01),the level of IL-2 in serum in model group were lower than that normal group(p＜0.01),the level of NO,COR,IL-6 and TNF in serum,ET and Angâ…¡in plasm in prevention group were higher than in normal group(p＜0.01),while they were obviously lower than in model group(p＜0.01),the level of IL-2 in serum in prevention group were lower than in normal group(p＜0.01),while they were obviously higher than that in model group(p＜0.01).Conclusion There was an obviously preventive effect of hepatocyte growth factor gene mediated by attenuated Salmonella on the stress ulcer model by water immersion and restraint stress in rats,typl-HGF can obviously improving the blood of stomatch,relief the condition of metabolic effect and oxygen deficiency;neuron-endocrine system to improve the regulation of the defense mechanism;can protect the integrity of stomach.The research result first step expresses that typl-HGF have a function of prevention and cure stress ulcer:the stomach and intestines hormone have an important function in the stress ulcer:on the whole the result showing that HPA play an important function during the occurrence of stress ulcer.This research takes stomach ulcer as to correspond point:passing to the normal group,model group,prevention group and control group,will produce or prevent from the stress ulcer.The meaning of this experiment lies that attenuated Salmonella is a favourable species of vehicle,commit a species of gene drug,will prevention and cure stress ulcer,give play to many kinds biology affect of HGF and the predominance of gene drug.
