objective To study the effect of phosphodiesterase isoenzymes5 inhibitor on the tissue of obstructive Ureteropelvic Junction and explore its significance.Method Twenty-four New-Zealand rabbits are randomly divided into three groups, the shamoperation group A(the right side is A1,the left side is A2), the right side juntion which was not intervened was seen as the normal control (A1)in group A ,the operation group B and the operation/drug group C.The Ureteropelvic Junction Obstrction(UPJO) model is established by partial obstruction of the juntion.Intravenous Pyelogram is done pre-or post-operation.HE stain and Microscopic examination were performed to observe the pathologic changes of the tissue. Immunohistochemical SP method was applied in testing the expression of nNOS and TGF beta-1 in the ureters.RT-PCR were applied to detect the mRNA expression of TGF beta-1.Result We observed more serious hydronephrosis in group B than group A and C,great pathologic changes were seen in group B but little changes in group C,no chang in group A.The level of TGF beta-1 in group B is significantly higer than group A and C(PAB<0.01,PBC<0.05),group C expressed more TGF beta-1 than group A,but in group B we detected less nNOS comparing to group A are C(PAB<0.01,PBC<0.05), and group C expressed less nNOS than group B(P<0.05).Conclusion More TGF beta-1 is expressed and nNOS decreased when the Juntion is obstructed , while,PDE5 inhibitor could reduce the expression of TGF beta-1 and increase the exprssion of nNOS.PDE5 inhibitor may keep the obstuction from developing by suppresion of TGF beta-1and up-regulation of nNOS.
