To study the activity of NOS(cNOS and iNOS) in cerebral ischemic reperfusion injury of rabbits and protection of Xuesaitong of neuronal nitric oxide synthase (nNOS), this experiment has studied, systematacially, activity change of NOS(cNOS and iNOS) in parietal lobe,frontal lobe, Hippocampus ,Cerebellum and medulla oblongata and protection of Xuesaitong in PL,FL,HIP,CB and MO in ischemic reperfusion injury of rabbits by SABC immunohistochemical method .The first experiment has studied the activity of NOS(cNOS and iNOS)of PL,FL,HIP,CB and MO with different times of cerebral ischemic reperfusion injury by metabolic chemistral method. The result revealed that the activity of cNOS(nNOS) begin to advance of 2h,reach to maxlmum about 6h, then begin to descent,24h,48h,72h continue to descent,retain to normal level eventually in parietal lobe,frontal lobe,Hippocampus ,Cerebellum and medulla oblongata of ischemic reperfusion injury in rabbits;but the activity of iNOS of PL,FL, HIP ,CB and MO with different times of ischemic reperfusion injury continue advance,72h group is the highest in all, the explain that NO of nNOS happen to effect in earlier period and iNOS happen to effect in advanced stage of ischemic reperfusion injury.Xuesaitong protection of in 2h group isn't obvious and there wasn't a significant change (P > 0.05).From 6h group to 72h group,the activity of healing obviously decrease with pathological group and close to normal level eventually,the activity of NOS always wander in surrounding of the normal.,the chage was significan(tP < 0.05). Comparing the healing groups with the normal,there wasn't a significant change(P > 0.05) The result also revealed that NO of nNOS happen to regenere effection in earlier period and iNOS happen to effect in advanced stage of cerebral ischemic reperfusion injury;the NOS activity of healing group always wander in surrounding of the normal.and Xuesaitong protect brain tissue of ischemic reperfusion injury. The second periment has studied Xuesaitong protection of positive neuron of nNOS in cerebrum and cerebellum with different times of cerebral ischemic reperfusion injury by SABC immunohistochemical method. The result showed that immune-positive neuron of nNOS cerebrum and cerebellum change in quantity ,average section area and first born tuber length with different times of cerebral ischemic reperfusion injury. Comparing the 2h and 6h groups of cerebral ischemic reperfusion injury with the normal , there wasn't a significant change (P > 0.05) in quantity,average section area and first born tuber length of positive neurons,but comparing the 24h,48h and 72h groups , quantity was obviously decrease(P <0.05),and average and first born tuber length in cerebrum and cerebellum was shorter and color of positive neurons become deep than with the normal and neurons of necrotic increase. Xuesaitong protection is not obvious in 2h group,it is thus evident that there wasn't a significant change (P>0.05)in quantityaverage section area,first born tuber length and brain tissue didn't obviously demerge. Healing groups of 6h,24h,48h and 72h, the quantity,average section area and first born tuber length of positive neuron of nNOS obviously increase with pathological group,nerve corpuscles of pyknosis and solution obviously decrease ,the quantity of dead positive neuron obviously decrease, there wasn't a significant change (P > 0.05).It is clear that Xuesaitong have protection of brain tissue in ischemic reperfusion injury. |
