Objective To trace refined distribution of neuron which control anterior palma skin in rat sympathetic chain and investigate the differency of calmodulin expression in the selective parts of rat sympathetic neuron and evaluate its relationship with the electrophysiological diversity of sympathetic postganglionic neuron.Methods (1) The experiment was performed in normal rat with the method of retrograde axonal transportation of horseradish peroxidase(HRP), by which HRP was inject in anterior palma subcutaneously. (2) The superior cervical ganglion, cervicothoracic ganglion and celiac ganglion of sympathetic chain in SD rats was collected. The mRNA and protein expression of Calmodulin were detected by RT-PCR and immunohistochemistry method respectively, the expression of mRNA and protein for calmodulin were compare among three ganglia.Results (1) Most of the labbled neuron distributed in the inferior cervical ganglion, cervicothoracic ganglion and third cervical ganglion. (2) Calmodulin were expressed in all the three choosen parts. The result of RT-PCR and that of immunohistochemistry corresponded with each other; There was no difference between the expression of calmodulin at the superior cervical and cervicothoracic ganglion(0.6631,3.5366, P﹥0.05). The expression of calmodulin in superior cervical and cervicothoracic ganglion was significantly higher than celiac ganglion(q=11.554,36.0912,10.9209,31.1996,P﹤0.05).Conclusion (1)Most of the labbled neuron distribute in the inferior cervical ganglion, cervicothoracic ganglion and third cervical ganglion, The ganglion which at higher location have more labbled neuron. (2) The differency of calmodulin expression in sympathetic postganglionic neuron suggest that it may play a role in the electrophysiological diversity of sympathetic neuron.
