| Objective:To explore expression patterns of lamininα1,α2,α3,α5,β1,β3,γ1,γ2 subunit in the human brain glioma and the relation between expression intensity of lamininα1,β1,γ2 and brain glioma invasion .Methods:â‘ To analyze the expression of lamininα1,α2,α3,α5,β1,β3,γ1,γ2 subunit in 77 brain glioma,8 normal brain tissue and 10 metastatic tumor specimens by immunohistochemistry and image analysis technique;â‘¡To analyze the relation between expression intensity of lamininα1,β1,γ2 and indirect tumor invasion exponent of 26 brain glioma specimens .Results:â‘ In the human brain glioma specimens,lamininα2,α3,β3 subunit staining was negative. Lamininα1,α5,β1,γ1,γ2 subunit positive staining was localized mainly in the kytoplasm and membrane of the gliomas cells,which intensity was correlated with pathological grade of glioma(P<0.001).â‘¡Lamininγ1,α5 subunit positive staining still was localized in blood vessel endothelial cells and basal membrane.The glioma and metastatic tumor groups were higher than the normal group in staining intensity of lamininγ1 which high expressed especially in proliferative endothelial and incrassate basal membrane.There showed the significant difference in expression between the low-grade(WHOâ… ,â…¡) and the high-grade (WHOâ…¢,â…£)glioma group(P<0.001),also showed the significant difference in expression between the metastatic tumor and the high-grade glioma group(P<0.01). Lamininα5 subunit positive staining intensity was not correlated with pathological grade in glioma blood vessel endothelial cells and basal membrane(P>0.05),but was stronger than in metastatic tumor group(P<0.05).â‘¢Lamininα1,γ2 subunit positive staining intensity in tumor cells was correlated with indirect tumor invasion exponent(P<0.001,P<0.01),and Lamininβ1 subunit positive staining intensity in tumor cells was not correlated with indirect tumor invasion exponent(P>0.05).Conclusion:â‘ The expression intensity of lamininα1,α5,β1,γ1,γ2 subunit in tumor cells was closely correlated with pathological grade of glioma.â‘¡The expression difference of lamininα5,γ1 subunit in brain glioma and metastatic tumor was probably correlated with the metabasis behavior of tumor cells.â‘¢The high expression of lamininα1,γ2 played probably a important part in the invasion of human brain glioma. |
