Research On Reducing Pyrogenous Reaction Of Glutaraldehyde Fixed Bovine Pericardium

Objective:Talk about the way to cut down the pyrogenicity of the bovine pericardium,and elevate its safety.To provide biomaterial which is much better in the mechanical character and safety for the body.In this article I try to reduce pyrogenous reaction of biological grafts by wash-out free glutaraldehyde from the tissue with alcohol solution.Methods:Select the bovine pericardium of the Qinchuan cattle,than to reject the adipose tissue. Cut out fifteen pieces of bovine pericardium with the size of 5cm×5cm.Deal these bovine pericardium 24 hours with glutaraldehyde solution in the density of 0.6%.Glutaraldehyde fixed bovine pericardium was immersed in each 50%,60%,70%,80%,90%alcohol solution for five minutes separately.Then the glutaraldehyde in each solution was determined by gas chromatography.Cut out fifteen pieces of bovine pericardium with the size of 10cm×10cm.Deal these bovine pericardium 24 hours with glutaraldehyde solution in the density of 0.6%.The glutaraldehyde fixed bovine pericardia washed in 60%,70%,90%alcohol solution were used for pyrogen test according to the method which is introduced in Chinese Pharmacopoeia 2005.No washed pericardium served as control.Results:All the datas were analysed by SPSS15.0.The glutaraldehyde remained in 60%and 70%alcohol solution(18.21±0.14‰and 11.91±0.09‰respectively,P＜0.05) was higher than in other groups(P＜0.01).The 60%alcohol post-treat pericardium cause the lowest pyrogen in rabbit model(0.5℃.0.6℃,0.8℃,0.9℃respectively,P＜0.05).Conclusion:60%alcohol solution effectively wash out free glutaraldehyde from bovine pericardium,and reduce the fever effect caused by the pericardium in rabbit model.
