Objective: 1. To evaluate the mutagenic toxicity of the bovine jugular vein grafts (BJVG) which are treated by Glutaraldehyde/Octanol , Polyepoxy Compound (PC) and Dye-mediated Photooxidation by the micronucleus (MN) test, chromosomal aberration (CA) test and Ames test; 2. To evaluate the systemic toxicities of these biomaterials by the pyrogen test, acute toxicity test, subacute toxicity test and chronic toxicity test.Methods: 1. Mutagenic toxicity: (a). The micronucleus test: The extracts of the treated BJVG and the controls were injected into the abdominal cavities of the mice. The injection was repeated every 24hs and totally 4 times, then after 24hs of the last injection the Giemsa-stained slides of the femoral bone marrow were made. The MN rates of the polychromosome erythrocytes (PCE) were scored and analyzed by SPSS 12.0. (b). The chromosomal aberration test: The injection of the extracts and the controls was the same as the micronucleus test. The Giemsa-stained slides of the femoral bone marrows cells were made 4hs after colchicine had been injected into the abdominal cavities of the mice, by the process of centrifuge-hypotonic treatment-fixation-smear-stain. The CA rates of the bone marrow cells were scored and analyzed by SPSS 12.0. (c). The Ames test: The extracts and the controls were added to the nutrient mediums in which the Salmonella typhimurium were inoculated, with the S-9 system added or not. The numbers of the strains in each culture dish were scored after 48hs of culture.2. Systemic toxicity: (a). The pyrogen test: The temperature was measured 3 times before, the mean temperature calculated, and 6 times after the extracts were injected into the veins of the rabbits. The maximal and total increase of the temperature of each rabbit were calculated, (b). The acute toxicity test: The extracts and the controls were injected into the veins of the mice and the degrees of the toxical effects were observed 4hs, 24hs, 48hs and 72hs later. (c).
