Objective Compare the effects ofα-linolenic acid and its anti-oxidation compounds with vitamin A,C,and E on the levels of total cholesterol,TC and triglyeride,TG in the serum of aged mice;on the anti-free oxidation damage:on the prolonging the life of flies and on the mRNA expression of the telomerase activity. To evaluate their effects on lowering-lipid, anti-oxidation damage and life prolonging.Methods(1) Aged mice used in the experiment were divided into 9 groups at random according to the different dosage.A control group of young mice was also used. The total cholesterol(TC) and triglyeride(TG) and high density lipoprotein(HDL) in the serum of the mice were measured. The serume-lipid lowering effect ofα-linolenic acid and its compounds with vitamin A,C,and E is evaluated.(2) Mice were divided into control groups of young and old.The aged group is further divided into 5 groups to be respectively treated withα-linolenic acid only,α-linolenic acid plus vitamin E,α-linolenic acid plus vitamin C,α-linolenic acid plus vitamin andα-linolenic acid plus vitamin ACE.Each group was consisted of 10 mice. The levels of superoxide dismutase(SOD) glutathione peroxidase(GSH-PX),and malondialdehyde (MDA) in serum of mice are measured to evaluate the effect of anti-oxidation damage.Fresh livers of mice were taken to measure the telomerase activity by the method of reverse transcription-polymerase chain raction(RT-PCR).(3) Flies are divided into 4 groups to be treated respectively withα-linolenic acid plus vitamin E,α-linolenic acid plus vitamin C,α-linolenic acid plus vitamin ACE and a control group.Every group was consisted of 50 flies. The average and the longest lifespan of every groups were record and analyzed. Results The values of TC,TG,and HDL in the serum of the mice in the experimental group were dramatically altered as compared with those of the control group. It was shown that the dosage of 1500 mg/kg ofα-linolenic,plus 40 mg/kg of vitamin E,8 mg/kg of vitamin C and 90 u/100g of vitamin A can significantly increase the SOD and GSH-PX,and decrease MDA of the mice serum;and obviousely prolong the lifespan of the flies. The above dosage can dramatically increase the mRNA expression of the telomerase activity, prolong the length of the telomerase and delay ageing. Conclusion The dosage of 1500mg/kg ofα-linolenic,plus 40mg/kg of vitamin E,8mg/kg of vitamin C and 90u/100g of vitamin A can significantly increase the SOD and GSH-PX and decrese serume lipid, prevent artery stenosis, delay ageing and prolong life.
