Using Cell Counting Kit-8 To Detect The Protection To HL60 Cells Preconditioned By Etomidate

Objective:This study obersive the protection to HL-60 Cells preconditioned by minidose etomidate.Methods:HL-60 cells were cultured at a concentration of 1.5×10~5/ml.Three groups were processed in accordance to different methods.The control group was blank(0μM etmidate for 24h);the preconditioning group was preconditioned with 1μM etomidate for 1h,washed etomidate out and allowed to recover for 4h,and then treated with 50μM,100μM,250μM,500μM,1000μM for 24h;The injury group was treated with 50μM,100μM,250μM,500μM,1000μM etomidate for 24h.Cell Counting kit- 8 measured the cell viability and proliferation.Results: Between the preconditioning group and the injury group the etomidate concentration inverse the HL60 survisingConclusions:1The cell counting kit-8 is saving time convenient and high sensibility.2,Etomidate can inhibit the viability of HL-60 cells,The minidose etomidate precondition can protect it.
