Objectives:To study the effect of erythropoietin rat kidne with delay fluid resuscitation after severe burns,and discuss the probable mechanism and protective effective of erythropoiet for rats kidney after delay fluid resuscitation after severe burns.Method:We took 45 healthy SD rats(provided by Kunming Medical College)to set up the models of delay fluid resuscitation after severe burns(TBSA30%),which were divided randomly into control group,delay fluid resuscitation after severe burns group and treatment group.Rats were subjected to 30%total boby surface area full thickness scald,followed by delay fluid resuscitation(40ml/Kg),EPO(3000IU/Kg)reatment group is going to inject by vena caudalis,30 min before delay fluid resuscitation group,We took one kidney tissue sample separately by 1,6,12,24h deIay after fluid resuscitation;examined and analyzed the kindey apoptosis with the methods of terminal -deoxynucleotidyl transferase mediated nick end labeling,(TUNEL), observed the pathological change of the kidney tissues of each groups,another kidney is to be examined SOD and MDA.at the same time we took blood by heart and examined BUN and Cr.Result:all 45 included rats were involved in the result analysis(1)treatment group compared with delay fluid resuscitation, BUN and Cr were lower significantly;(2)compared with delay fluid resuscitat ion MDA less but SOD more significantly;(3)compared with delay fluid resuscitation renal histologic injury were obviously attenuated by EPO in EPO treatment group.renal tubular epithelial cells and apoptosis of kidney tissues of lightly swelled,basement membrane wasmostly integrated.There were few casts in renal tubular.Conclusion:The EPO treatment can lessen kidney of delay fluid resuscitation injury,the mechanism and protective effective of which is probably through preventing oxygen free radical damage and elevating endogenouns antioxidation potential and to lessen apoptosis of kidney tissues.
