Objective: To analyze the HLA-DRB1 allele genetic susceptibility to Systemic scleroderma patients of Guangxi region. And explore the association between HLA-DRB1 genetic polymorphism and systemic scleroderma , further analyze the immunological mechanism of systemic scleroderma.Methods: Thirty-three patients (18 women and 13 men ; mean age at diagnosis 44.7 years) fulfilling the American College of Rheumatology (ACR) classification criteria for SSc were enrolled. The controls were 40 healthy, disrase-free Chineses . HLA-DRB1 genotypes were assessed by the polymerase chain reaction- special sequence primers(PCR-SSP) method.Results : 1.The distribution of HLA-DRB1 alleles:(1) 6 alleles were observed in systemic scleroderma group. The range of allelic frequencies is from 1.52% to 31.82%. Among them the DRB1*15 allele is popular (31.82%), the DRB1*13 is rare (1.52%), DRB1*01 and *07 have not been examined.(2) 8 alleles were observed in control group. The range of allelic frequencies is from 15.00% to 1.25%. Among them the DRB1*15 ,*16 allele are popular(15%,10%), the DRBl*01,*04,*08,*ll,*13 are rare(1.25% ,equally).2. The comparison of HLA-DRB1 allelic frequency:(1) DRB1*15 allelic in the systemic scleroderma group was significantly higher than that in the control group (31.82%VS15.00%,X~2 =8.259, RR= 4.0833, P<0.01) .(2) No significant difference of other allelic frequencies was observed between the two subjects.Conclusion: HLA-DRB1*15 is closely related to the invasion of systemic scleroderma in Guangxi ;HLA-DRB1*15 may be the susceptible gene of systemic scleroderma in Guangxi; DRB1*01 and *07 have not been examined .
