Objective: To explore the potential association between HLA-DQA1 and DQB1 Alleles with systemic scleroderma (SSc) of the Zhuang Nationality and Han Nationality in Guangxi region.Methods: Polymerase chain reaction-special sequence primers(PCR-SSP) was used to study the HLA-DQA1 and DQB1 alleles in 50 Zhuang Nationalty patients with SSc and 100 healthy controls of the Zhuang Nationalty and 50 Han Nationalty patients with SSc and 100 healthy controls of the Han Nationalty in Guangxi Province.Results : 1. The distribution of HLA-DQA1 alleles(1) 8 alleles were observed in systemic sclerosis group In Zhuang nationality, the range of allelic frequencies is from 0.000 to 0.300. Among them the DQA1*0102,*0401 allele are high (0.210,0.300), the DQA1*0103, *0301is rare (0.040,0.050), and the DQA1*0201, *0302 have not been examined.In controls, 9 alleles were observed .the range of allelic frequencies is from 0.000to 0.215.Among them the DQA1*0102 allele are high (0.215), the DQA1*0103,*0301is rare (0.050,0.055), and the DQA1*0201,*0302 have not been examined. (2) 8 alleles were observed in systemic sclerosis group In Han nationality, the range of allelic frequencies is from 0.000 to 0.190. Among them the DQA1*0101,*0104,*0601 allele are high (0.150, 0.190, 0.170), the DQA1*0103, *0301, *0401is rare (0.070, 0.080, 0.080), and the DQA1*0201,*0302 have not been examined. In controls, 9 alleles were observed .the range of allelic frequencies is from 0.000 to 0.225. Among them the DQA1*0101, *0102 allele are high (0.200, 0.225), the DQA1*0103, *0301, *0601 are rare (0.060, 0.070, 0.030), and the DQA1*0302 has not been examined.2.The distribution of HLA-DQB1 alleles(1) 4 alleles were observed in systemic sclerosis group In Zhuang nationality, the range of allelic frequencies is from 0.000 to 0.190. Among them the DQB1 *0301, *0501,*0601 allele are high (0.150,0.120,0.190), the DQB1*0201is rare (0.080), and the DQB1*0401 have not been examined.In controls, 4 alleles were observed .the range of allelic frequencies is from 0.000to 0.140. Among them the frequencies of DQB1*0301 allele is high (0.140), the DQB1*0501,*0601 is rare (0.040,0.075), and the DQB1*0401 has not been examined.(2) 4 alleles were observed in systemic sclerosis group In Han nationality, the range of allelic frequencies is from 0.000 to 0.180. Among them the DQB1*0601 allele are high (0.180), the DQB1*0201,*0501is rare (0.070, 0.050), and the DQB1*0401 have not been examined.In controls, 4 alleles were observed .the range of allelic frequencies is from 0.000 to 0.135. Among them the frequencies of DQB1*0201 allele is higher (0.135), the DQB1*0601 is rare (0.065), and the DQB1*0401 has not been examined.3. The comparison of HLA-DQA1-DQB1 allelic frequency:(1) The allele frequencies of HLA-DQA1*0401 and DQB1*0501 and DQB1*0601 were significantly increased in Zhuang Nationalty patients with SSc than those of the controls((0.300 VS 0.135,RR=4.056,χ~2=15.407,Pc=0.001; 0.120 VS 0. 040,RR=4.472,χ~2=10.653,Pc=0.004; 0.190 VS 0.075,RR=3.473,χ~2=10.06,Pc=0.008 ).(2) The allele frequencies of HLA-DQA1*0401, and DQA1*0601and DQB1*0601 were significantly increased in Han Nationalty patients with SSc than those of the controls((0.080 VS 0.010,RR=9.333,χ~2=8.371,Pc=0.036; 0.170 VS 0.030,RR=8.071,χ~2=20.130,Pc=0.000; 0.180 VS 0.065,RR=3.764,χ~2=10.755,Pc=0.004 )(3) The allelic frequencies of HLA-DQA1*0201 was significantly less in Zhuang Nationality and Han Nationality patients with SSc than those in the control(0.000 VS 0.115,χ~2=13.583,Pc=0.002 and 0.000 VS 0.105,χ~2=12.209,Pc=0.004).(4)No significant difference of the other allelic frequencies was observed between the two subjects.Conclusion: It suggested that the HLA-DQA1*0401 and DQB1*0601 may be the susceptible genes for SSc in Guang Xi Zhuang Nationality and Han Nationality,and HLA-DQB1*0501 may be the susceptible genes for SSc in Guang Xi Zhuang Nationality ,and HLA-DQA1*0601may be the susceptible genes for SSc in GuangXi Han Nationality. While the HLA-DQA1*0201 alleles may be protecting gene for SSc in GuangXi Zhuang Nationality and Han Nationality. |
