Establishment And Comparison Between Two Rats Models: CIA And PIA

Objective:In order to get a better animal model for the basic scientific research on rheumatoid arthritis,we have established two Wistar rats models,i.e.,PIA and CIA,and compared their differences on clinical features,laboratory examination and pathological characteristic.Method:We have randomly divided 60 rats into 3 groups,CIA,PIA and control group,20 for each one,and established CIA rats model by intradermally injecting collageⅡacid solution 0.01ml/g into the rat's back or the bottom of rat's tail,PIA rats model by intradermally injecting pristine 0.003ml/g into the rats' back or the bottom of rat's tail as well as rats of control group is injected isotonic Na chloride.After that,we observe the clinical symptom of all the three group rats,the degree of the rats' arthritis base on the joint score and pathological characteristic so as to sure whether the model is successful.And then,we compare the features during the course of disease,immunological index of serum(have tested anti-CCP antibody and RF in the blood serum of the rats),pathological characteristic and model successful rates between successful PIA rats model and CIA rats model.Results:(1)The rats of CIA and PIA both have symptom of arthritis,the pathology of which is rodent synovitis with synovial hyperplasia,many of inflammatory cells infiltration,cartilage even bone lost occurring.(2)The model successful rate of CIA rats model is 65%,higher than that of PIA rats model of 30%,which is discrepant in statistic(P＜0.05).(3)CIA rats' arthritis appears earlier and more serious than PIA.Furthermore,CIA rats' pathological changes are more serious than that of PIA with its cartilage destroyed.Arthritis symptom of CIA rats started easing 3 weeks later after it appeared and no more serious arthritis.Nevertheless,the arthritis symptom of PIA rats always appeared repeatedly,which is more similar to the rheumatoid arthritis of human beings.(4) In PIA group,RF tite of some serum slightly elevates when RF tite of CIA group is completely normal.By the same time,the variance of RF tite average between 2 groups is meaningful on statistics.Anti-CCP antibody of two groups are both normal.Conclusion:(1)Both CIA rats model and PIA rats model can be successfully established with inbred line Wistar rats.(2)CIA rats model established with inbred line Wistar rats is more practical than that with PIA rat model.
