Objective To observe the function and mechanism of oxymatrine treat rheumatoid arthritis in rats with collagen-induced arthritis(collagen-induced arthritis, CIA), investigate imbalance between Treg and Th17 cells. To observe the effect of oxymatrine on joint swelling and arthritis index, the influence of joint pathological changes of CIA rats, the level of serum IL-17 A and TNF-α, and its effect on Foxp3 m RNA and RORγt m RNA and the protein expression of spleen cells. To explore the effect of oxymatrine on rheumatoid arthritis in rats, and the possible molecular mechanisms of immunity. At the same time, to provide a theoretical basis and experimental evidence for the clinical treatment of rheumatoid arthritis.Methods 60 SD rats, weighing 200 ± 20 g, 10 were randomly selected as a control group, and the remaining 50 for the experimental group were randomly divided into five groups, namely: the model group, positive drug group(dexamethasone group), high dose group of oxymatrine, medium dose group and low-dose group,each group has 10 rats.(1) Establishment of rat model of CIA: 50 SD rats in the experimental group were constructed CIA model in SD rats by subcutaneous injection of hybrid emulsion into right toe 0.1m L, after a week, strengthen immunity.The abdominal cavity injection of different concentrations of oxymatrine and dexamethasone treatment from the 8th day;(2) HE staining: SD rats were sacrificed at 43 day, take each rat knee and ankle fixation, embedding, sectioning by HE staining,observe the changes of articular synovial hyperplasia and inflammatory changes in the joint synovial tissues;(3) To detect the level of serum IL-17 A and TNF-α by ELISA.(4) To measure the m RNA of Foxp3 and RORγt in spleen cells by RT-PCR technique.(5) To detect protein expression of Foxp3 and RORγt t by Western-blot.Results(1) Oxymatrine relieved paw swelling and arthritis index of CIA rat significantly(P<0.05).(2) Oxymatrine reduced the synovial hyperplasia of CIA rats.(3) Oxymatrine down-regulated the level of TNF-α and IL-17 A in serum of CIA rats(P<0.05).(4) Oxymatrine inhibited the level of RORγt m RNA and promoted level of Foxp3 m RNA.(5) Oxymatrine inhibited the expression of RORγt and promoted the expression of Foxp3.Conclusion(1) Oxymatrine has the effect on anti-rheumatoid arthritis.(2) Oxymatrine could play an anti-arthritis effect by regulate imbalance between Treg and Th17 cells.
