Malignant tumor is a dease which threaten the haalth of the human,so it is important to find the the tumor therapy and antitumor medcine.It is thinked that the heat toxin is a main pathological factors of the malignant tumor by TCM,the weak and heat toxin is the pathological basis of tumor.The key pathogenesis is accumulation of heat toxin and phlegm-dampness or water and dampness.Our previous study.have find that Herba Hedyotis Diffusae can killed cancer cell by inducing the HSP70 expression in H22 liver cancer cells and the reinforcement of its immunogenicity.By studing the influence of HSP70 expression induced by Herba Hedyotidis Diffusae on the pathological changes and T lymphocyte subgroup in mice with H22 cells transplanted tumor.we know that H22 liver cancer cells pretreated by HHD can increase mice active immunity on the same kind of transplanted tumor.The machnism may be related to the induction of the HSP70 expression in H22 liver cancer cells and the reinforcement of its immunogenicity.We have knowed that HSP70 is higher expressed in the tumor,specially in the malignant tumor these yaars with more study of the HSP70.So by HSP70 we can research the tumao,especially in the study of the tumor immunity,the main idear is that HSP70 can take tumor antigen peptides which is called HSP70-PC to causes the non-specific immune response to the tumor cell by presenting tumor polypeptide to the T lymphocyte cell.In recent years,with the development and application of molecular biotechnology in the tumornoliogy,we know that tumor generation have relation with the change of the cell gene.P16 is a tumor suppressor gene which is findec in these years.p16 combined cyclin-dependent kinase cyclin-dependent kinase4(CDK4)to prevent cell growth from G1 phase to S phase and inhibits its growth t.The abnormal of the P16 gene can cause tumor.so we can study the mechanism of the tumor by p16.Aim:To study the effect on the HSP70 and P16 expression induced by Herba Hedyotidis Diffusae(HHD)on H22 Cells Transplanted Tumor and theantitumor mechanism of Herba Hedyotidis Diffusae(HHD)Methods:Mice with peritoneal transplantation of H22 liver cancer cells were treated with Herba Hedyotidis Diffusae(HHD)and Radix Codonopsis(RC)by gastric gavage respectively for 12 days.Then H22 cells isolated from the mice treated by herbal medicine were subcutaneously injected into mice right axilla.The mice immunized by right subaxillary injection were divided into 5 groups: HHD group(immunized with HHD-treated H22 cells),blocked HHD group(immunized with HHD-treated H22 cells blocked by HSP70 monoclonal antibody),RC group (immunized with RC-treated H22 cells),blocked RC group(immunized with RC -treated H22 cells blocked by HSP70 monoclonal antibody)and blank control group (immunized with RPMI-1640).The pathologic changes of tumor tissue,thymus gland and spleen,as well as the expression of peripheral blood CD4+and CD8+ in the mice were observed.And The in-vivo expression of HSP70 in ascitic H22 cells of different groups was detected by immunohistochemical method.Results:As compared to the blank control group,CD4+ and CD8 + expression in HHD group and blocked HHD group were up-regulated obviously,CD4 + and CD8 + expression in RC group and blocked RC group were increased also,but the difference was insignificant.1,compared to the blank control group,the HSP70 expression in blocked HHD group and blocked RC group were up-regulated obviously.The difference of HSP70 expression between blocked HHD group and blocked RC group is significant(p<0.05).2,compared to the blank control group,the HP expression in HHD group,blocked HHD group,RC group and blocked RC group were up-regulated obviously,the HP expression in HHD group and blocked HHD group is higher than RC group and blocked RC group(p<0.05) Conclusion:HHD exerts some inhibitory-killing effect on living murine H22 cells,and its mechanism may be related to the increase of immune function and to the cytotoxic effect.
