Experimental Study Of Anti-asthmatic Action In Sorbus Tianshanica Ruper

Objective:To study the anti-asthmatic action of Sorbus tianshanica Ruper Which is a special plant in xingjiang and study its preliminarily mechanism.Content and Method:to Observe the antagonist effect of Sorbus tianshanica Ruper against histamine (HA) and acetylcholine(Ach) both in wholes and in vitro:With the experimental method of isolated guinea pigs tracheal smooth spiral muscles,recorded the dose-response relationship curve,compared the maximum contract response extent of isolated guinea pigs tracheal smooth spiral muscles before and after treated by Sorbus tianshanica Rupe; Asthmatic model of guinea pigs were established by aerosol inhalation of mixed solution of histamine and acetylcholine for the observation of effects of Sorbus tianshanica Ruper on the latent period of asthma attack;observed the effect of Sorbus tianshanica Ruper on inflammatory mediator and inflammation cell of asthmatic model of mice:The delayed allgergic inflammation model in mice was induced by OVA,blood collection from eyeball,differential blood count,Bronchoalveolar lavage fluid(BALF) collected,EOS number counted,tested the nitric oxide(NO) in the supernatant of BALF;With the inflammation mice ear due to xylene,observed the reducing extent of inflammation reaction after treated by Sorbus tianshanica Ruper;used the method of ammonia water causing cough to observe the change of cough times in mice before and after treated by Sorbus tianshanica Rupe.Results:Sorbus tianshanica Rupe prevented the attack of athma caused by HA/Ach,delayed the latent period of asthma attack,Sorbus tianshanica Rupe decreased maximum contract response extent of isolated guinea pigs tracheal smooth spiral muscles,Sorbus tianshanica Rupe had the effect of antagonismin the delayed allgergic inflammation model in mice which was induced by OVA,Sorbus tianshanica Rupe reduced the number of EOS in BALF and peripheral blood of inflammation mice,decreased the content of NO which is a damage medium,Sorbus tianshanica Rupe(5,10,20g/kg) relieved cough in mice caused by aerosol inhalation of ammonia water.Conclusion:Sorbus tianshanica Rupe water extract inhibited the contract and spasm in isolated guinea pigs tracheal smooth muscles;antagonized the attack of athma in asthmatic model of guinea pigs caused by HA/Ach,It also had the effect of antagonism in the delayed allgergic inflammation model in mice which was induced by OVA and the effects of anti-inflammation,relieving cough,we concluded that its possible mechanism of action as follows;inhibition of the formation and activation of inflammatory mediators;inhibition of the chemotaxis and aggregation of inflammatory cells;modulation of immune system.
