Objective: Using the modified rabbit in situ lung preservation model to investigate the protection of Edaravone for the lungs in the pulmonary ischemical reperfusion under the lung transplantation models.Method: Selection 20 adult rabbits,each Weigh 2.5-3.0Kg.(Provided by the animal experiments center of the Faculty of Medicine of Jiangsu University).The rabbits were divided into two groups (LPD group and Edaravone group)randomly,10 in each group. To establish in situ lung preservation model, In LPD group,the lungs were perfused and preserved with LPD solution,while in Edaravone group,the lungs were with LPD+Edaravone solution which add Edaravone 0.05 mg/mL into LPD solution. Each group got the blood samples at the same time. Before ischemia,reperfusion 1h,2h for PVO2 Before ischemia,reperfusion 30min,60min,90min,120min for MDA,SOD,iNOS,eNOS; At the end of the operation got the lung tissue for lung wet/dry weight ratio and examined by the pathology.Results:1 .The comparison of the partial pulmonary vein oxygen pressure after the reperfusion: The PVO2 all decreased after reperfusion in two groups,but it was higher in Edaravone group than that in LPD group in the same time. It was significant(P < 0.05). 2.MDA measurement, SOD expression: The MDA all increased after reperfusion in two groups,but it was lower in Edaravone group than that in LPD group in the same time; The SOD expression all decreased after reperfusion in two groups,but it was higher in Edaravone group than that in LPD group in the same time. The comparison of MDA measurement, SOD expression in LPD group and Edaravone group has significance (P < 0.05) after reperfusion.3. iNOS,eNOS expression: The iNOS expression all increased after reperfusion in two groups,but it was lower in Edaravone group than that in LPD group in the same time; The eNOS expression all decreased after reperfusion in two groups,but it was higher in Edaravone group than that in LPD group in the same time. The result of two groups has significance(P < 0.05)after reperfusion.4. W/D of lung tissue: the comparison of the lung tissue W/D in LPD group and Edaravone group shows significance (P < 0.05) .5. The pathological changes in lung tissue through optical microscope:In the LPD group,a part of alveolus pitted fall, alveolar spetum is widened, accompanied by a large number of inflammatory cells,micro-vessel is exaggerated, vascular endothelial is edema.But in Edaravone group, the configuration of alveolus is clear, vascular endothelial is integrity, a few of inflammatory cells soakaged in alveolar spetum,small vessel exaggerated and congested. 6.The pathological changes in lung tissue through electron microscope:In the LPD group, the alveolar septa is obvious edema, the RBC leakage in alveolar cavity is obvious, deposited in piles,inflammatory cells infiltrate seriously, The lamellar body in type II alveolar epithelial cell cytoplasm was emptied,microvili were defluxion,endothelial cell swelled obviously;But in Edaravone group most of the alveolar organizational structure is normal,type II alveolar epithelial cells are generally normal, The surface microvilli is still visible,lamellar body ablated partly, only a few of RBC and inflammatory cells coulde be seen in the alveolar cavity, endothelial cell is basically nomal.Conclusion: In the models of rabbit in situ lung preservation, Edaravone can elevate the OFR clearing ability of the organism and effectively reduce the ischemical reperfusion injury of pulmonary grafts, and improve the early pulmonary function after the transplantation. Use the Edaravone as a additive is effective.
