| Objective: Explore the damage of lung tissue induced by intermittent hypoxia and discuss the protective effect and mechanism of edaravone on lung tissue injury mediated by intermittent hypoxia.Methods: Divide 36 healthy SD male mice into 4 groups randomly and evenly:normal control group(group A),intermittent hypoxia group(group B),intermittent hypoxia+edaravone group(group C)and intermittent hypoxia+normal saline group(group D).Use intermittent hypoxia tank to establish intermittent hypoxia mice model for 8 weeks,then take lung tissue of mice to observe morphological changes in lung tissue with HE staining and Masson staining,detect the level of MDA content and the activity of SOD and GSH-Px with manual biochemical method,detect the level of L-6and β-catenin expression with immunohistochemical,detect level of β-catenin m RNA with RT-PCR.Result:(1)HE staining: There was no obvious morphology abnormality in the lung tissue of the mice in group A.The main morphological changes of the lung tissues of the mice in group B and D were partial alveolar dilatation,lymphocyte infiltration in the interstitium,and thickening of alveolar septum.Compared with group B and D,the above-mentioned morphological changes of lung tissue in group C were reduced.(2)Masson staining: Compared with group A,the blue stained area of group B and D increased significantly,that is,more collagen fibers were deposited;compared with group C,the blue stained area of lung tissue in group B and D increased,that is,more collagen fibers were deposited.(3)MDA,SOD,GSH-Px:Compared with group A,the level of MDA content in group B and D increased,the activity of SOD and GSH-Px decreased;compared with group C,the level of MDA content in group B and D increased,and the activity of SOD and GSH-Px decreased.(4)Immunohistochemical: Compared with group A,in the lung tissue of group B and D,the level of IL-6 and β-catenin expression increased.Compared with group C,in lung tissue of group B and D,the level of IL-6 and β-catenin expression increased.(5)RT-PCR to detect the level of β-catenin m RNA: Compared with group A,β-catenin m RNA expression increased in lung tissue of group B and D.Compared with group C,β-catenin m RNA expression upregulated in lung tissue of group B and C.Statistical analysis of the above data shows that the P values are all less than 0.01,and the differences have statistically significant.Conclusion:1.Intermittent hypoxia promotes inflammatory response and inflammatory cytokine release through oxidative stress to cause lung tissue damage,and mediates the deposition of collagen fibers in lung tissue by up-regulating the expression of β-catenin protein probably;2.Edaravone may inhibit oxidative stress,reduce inflammation,inhibit the expression of β-catenin protein,and then protect the lung tissue from damage induced by intermittent hypoxia. |
