PrefaceSilicosis is a pulmonary disease contracted through the inhalation of high concentrations crystalline silica,which induces a fibrogenic tissue response.It endangers severity to the human body,and we still has no valid of treatment means. There has been researches suggesting that some cytokines and the network system it forms play a crucial role in the occurrence and the development of the lung fiber turn. Transfer growth factor beta 1,platelet derived growth factor and conective tissue growth factor etc play prominent roles in lung fibroblast disintegrating and propagating and the collagen synthesizing and degradating.The studies were mostly vitro,and it may prove the network relationship of the cytokines,but may not necessarily match the vivo condition.Then,the serum contents of TGF-β1,PDGF and CTGF in the 58 patients with silicosis were detected and analyzed by means of enzyme linked immunosorbent assay.To explicit TGF-β1,PDGF,CTGF serum expression significance in patient with silicosis,and explore the function in the mechanism and correlation among them.MethodsFifty-eight patients with silicosis come from the ninth hospital of ShenYang from December 2006 to March 2007 and aged 61~85 years,tweny~nine healthy persons were in the control group and aged 59~83 years.All selected participant underwent routine blood test,routine urine test,liver function,kidney function and Chest X-ray inspection.The serum levels of TGF-β1,PDGF,CTGF were measured by Enzyme-linked Immunosorbent Assay(ELISA). ResultsThe serum levels of TGF-β1,PDGF,CTGF in patients with silicosis were 8.12±1.17ng/ml,942.62±306.89pg/ml,82.24±44.28pg/ml privately,and the serum levels of control group were 7.04±1.32ng/ml,731.28±288.06pg/ml,76.67±39.28pg/ml. There were significantly statistical differences between the case group and the control group in the serum levels of TGF-β1 and PDGF(p<0.05).There was no significantly statistical difference between the two groups in the serum levels of CTGF(p>0.05). The serum levels of TGF-β1 and PDGF in the patients with silicosis decreased with the silicosis stage going up(p<0.05).There was the bivariate correlation not only between the serum levels of TGF-β1 and PDGF(r=0.437,p<0.05)in the whole objects but also the serum levels of YGF-β1 and CTGF(r=0.469,p<0.05).ConclusionsThe expression ofTGF-β1,PDGF and CTGF as well as its mutual regulation is an important stage of silicosis pathogenesis.The expression levels of TGF-β1 and PDGF increase with the stages of silicosis,and tightly correlate with the extent of silicosis.
