AIM::The study was to investigate the effect of Epimedium on calcium and phosphorus metabolism and contents of IL-1β,IL-6,TNF-a in degenerated cervical vertebral of rats,to observe the inhibition effect of Epimedium in bone and disc of degenerated cervical vertebral and to explore the mechanism of Epimedium for the prevention and treatment on Cervical Spondylosis.METHODS:Thirty 6-month-old SD rats,weighing(220±20)g each,were randomly divided into two groups,model groups and sham group(Group C),male and female informality.The animal models were established by removing the muscle groups,the supraspinal and interspinal ligaments in the neck of rats.And cervical vertebrae X-ray were conducted at the time of 1 month and 3 months.After 3 months the animal models were established successfully according to X-ray result verify,they were divided into 2 groups randomly,Epimedium model group(Group A)and Saline model group(Group B).Then Group A and Group B were administered with Epimedium and Saline respectively,Group C were administered with Saline every day.The rats were killed to collect experimental samples after 2 months treatment,and then the blood samples were undergone for testing the contents of calcium,phosphorus,alkaline phosphatase and cervical bone specimens for testing calcium,phosphorus by using automatic biochemical analyzer respectively.And the cervical intervertebral discs specimens were to test the contents of IL-1β,IL-6 and TNF-αin each specimen by means of Radioimmunoassay. RESULTS:①Afterl month experiment,the x-ray suggestion had no significant variation between the model group and the normal control group(P>0.05).However,3 months later,x-ray photograph after model establishment showed that the physiological curvature disappeared or even recurved,intervertebral foramen became smaller, intervertebral space became narrow and osteophyte was formed in vertebrae.There was significant difference between model groups and control group(p<0.05).X-ray photograph's Scores of Model group at 1 month and 3 month have significantly different(P<0.01).②At the end of the experiment,the serum phosphate and alkaline phosphatase level in Group A were higher than that of the other groups(P<0.01),while there were no difference in their serum's calcium levels(P>0.05).③The cervical vertebrae bone calcium and phosphate levels in Group B were lower than that of the other groups(P<0.05).The cervical vertebrae bone calcium and phosphate levels in Group A were higher than that of the other groups(P<0.01),but the cervical vertebrae bone contents of calcium there were no difference between Group A and Group B.④The contents of IL-1β,IL-6 and TNF-αin discs of degenerated cervical vertebral rats model(Group B)were significantly higher than that of normal rats(Group C).The contents of IL- 1β,IL-6,TNF-αin disc of Epimedium model group(group A) were lower than that of Group B,but it had no difference than that of Group C.CONCLUSION:(1)The degenerated cervical vertebral in the rats can be established successfully through resection of the cervical supraspinous and interspinous ligaments from the cervical spine elicited by surgery after 3 months.(2) Epimedium can improve the Contents of calcium and phosphorus in degenerated cervical vertebral as well as prevent the degeneration of cervical bone.(3) Epimedium can increase the activity of the serum's alkaline phosphatase in degenerated cervical vertebrae,and it might inhibit the formation of osteophyte and delay cervical bone degeneration. (4)Epimedium can decrease the contents of IL1-β,IL-6 and TNF-a in disc of degenerated cervical vertebral and it might inhibit the formation of inflammatory effect,Which might play an important role on the inhibition of the intervertebral discs degeneration.(5)The reduction of cervical vertebral calcium and phosphorus was the pathological results after degenerated cervical vertebrae.(6) The cytokines which was released by degenerated cervical vertebral disc can accelerate disc degeneration.(7)The contents of IL-1β,IL-6,TNF-αincreased in the degenerated cervical vertebral disc and levels of calcium,phosphorus in the degenerated cervical vertebral bone decreased may be an important factor for Cervical Spondylosis.
