| The Soybean Minor Peptides are the products that the soy protein hydrolyzed. They're easily to be digested and absorbed, can provide the energy rapidly to organism. As active polypeptides, they have many physiological functions, they can resist radiation, decrease the blood pressure and blood fat and promote fat metabolism. They're the ideal functional foods. For the better development and application of Soybean Minor Peptides, we study and discuss their anti-fatigue effects and mechanisms by the fatigue mice model. In this work, the fatigue mice blood lactic acid, glycogen, SOD activity, immune function and other biochemical index were detected.Methods and Results:1. Soybean Minor Peptides were prepared by the new extraction technique from defatted soy flakes hydrolysized by enzymatic method .In this study,soybean minor peptides were obtained from the pretreatment and hydrolyzing by crude complex trypsinase and neutral proteinase hydrolysis and purification from defatted soybean flakes.The molecular weight distribution of Soybean Minor Peptides analyzed by the chromatography. The range is between 300-700 Da.2. Anti-fatigue effect of Soybean Minor Peptides were through the exhaustive time of the mice heavy loaded swimming in the flume by prepared mice fatigue model. The time that mice enter the water is called the exhaustive time. It can be observed mice exhaustive swimming time, the experimental group showed that swimming time respectively were 39.7±2.2 min, 50.2±3.4 min and 52.5±3.1 min. The results show the exhaustive swimming times of high-dose group mice were longer than the control group 41.4±1.8 min. It indicated that Soybean Minor Peptides have the anti-fatigue effect.3. Soybean Minor Peptides effect on the fatigue mice's serum urea nitrogen Through Diacetyl monxime's reagent colorimetic method to analyse the serum urea nitrogen of mice, it is showed that in the low, middle and high dose experimental mice groups, serum urea nitrogen content were 20.27±1.79 mg / ml, 20.35±1.86mg/ml and 20.40±2.07mg/ml, compared with the control group 20.30±1.77mg/ml serum urea nitrogen had no significant change.4. Soybean Minor Peptides effect on the fatigue mice's liver glycogen effect were through antrone Chromametry method, it is showed that in low, middle and high dose groups, the liver glycogen content of fatigue experimental mice are 4.09±0.77%, 4.68±0.83% and 5.37±0.90%. And the glycogen content of the high-dose group are more than control group 4.58±0.82%, showing that Soybean Minor Peptides can promote fatigue mice liver glycogen content, thus plays the anti- fatigue role. 5. Soybean Minor Peptides effect on the fatigue mice's blood lactic acid were through spectrophotometric analysis method, it show that fatigue experimental mice blood lactate levels are 47.84±1.76,48.79±1.97 and 37.45±1.06 in low, middle and high dose group, the high dose group decrease significantly compared to the control group, 49.84±1.79, suggesting that Soybean Minor Peptides affect fatigue mice can be achieved through reduction of blood lactate.6. Soybean Minor Peptides effect on the activity of the red blood cells and super-oxide dismutase (SOD) of liver tissue cell Through the autoxidation of Pyrogallol methods,it is showed that low, middle and high dose experimental mice SOD activity of red blood cells were 14.37±1.24 U / mg, 21.01±1.74 U / mg and 21.85±2.15 U / mg, compared with the control group 14.72±0.75 U / mg, the high dose experimental mice SOD activity were significantly increased. And it is also showed that low, middle and high dose experimental mice SOD activity of the liver tissue cell were 12.38±1.69 U / mg, 13.14±1.97 U / mg and 13.58±1.58 U / mg, compared with the control group 12.70±1.36 U / mg, the middle and high dose experimental mice SOD activity increased the tendency slightly, suggesting that Soybean Minor Peptides can play a role in anti-fatigue by inhibit the activity of SOD and increase the release of oxygen free radicals.7. Soybean Minor Peptides effect on proliferation of T lymphocyte The result of T lymphocyte transformation by MTT show that Soybean Minor Peptides can prompt proliferation of mice T cell, suggesting that Soybean Minor Peptides can improve the cellular immune function.8. Soybean Minor Peptides effect on mice macrophage phagocytosis By analyzing the experimental results of peritoneal macrophage phagocytosis swallow chicken erythrocytes, it is showed that Soybean Minor Peptides can significantly increase the percentage of macrophage phagocytosis swallowed of erythrocytes, suggesting that Soybean Minor Peptides can promote macrophage phagocytosis.We have prepared Soybean Minor Peptides successfully and confirmed Soybean Minor Peptides have the anti- fatigue function through the mice fatigue model. Through the biochemistry method study, it is suggested that Soybean Minor Peptides may enhance the liver glycogen reserve, reduce the blood lactic acid content, strengthen the SOD activeness, repress release of the oxygen free radical and improve the cellular immune function to play the anti- fatigue role. |
