Object: The inflammation factor(IL-1β) was used to induce rat Osteoarthritis, then observing expression of TGF-βat synovial membrane and cartilage in rat Osteoarthritis at intervals.Methods: 45 healthy rats were chosed in this experiment. The mould was divided into randomly in 3 group: group A (high dosage group), group B (low dosage group), group C (control group), each group with 15 rats. The IL-1β(10ng/ml) with 0.1ml was injected gastrocnemiusm of group A daily, the group B with IL-1β(1ng/ml) and the group C with same dosage saline per day. On 2, 4 and 8 weeks the rats were killed and knee joint was selected, first the specimen was immersed in 4% Formaldehyde, second transfered into Decalcification fluid, then buried in paraffin, the last the specimen were cutted with 4~5μm section. HE Stain and Ultra-Sensitive SP Immuno- histochemistry were chosed for observing expression of TGF-βin knee joint.Results: A little of TGF-βwas found in the intra-cell of the control group, the immuno-histochemical staining reaction was positive at 2 weeks, At the end of the 4 week the concentration of TGF-βreached the highest level, Until 8 weeks the positive cells declined dramatically in experiment group. But in experiment, the TGF-βdosage of expression in high dosage group were large obviously the low group.Conclusion: It is concluded that TGF-βmay play a dominant role in bone plerosis and be released at intervals. Moreover, the TGF-βdosage of expression in high dosage group were large obviously the low group.
