Objective To investigate the effects of mycophenolate mofetil(MMF) on matrix metalloproteinase-9(MMP-9) and tissue inhibitors of metalloproteinase-1(TIMP-1) expression in diabetic rats renal tissues.Methods Male Wistar rats were randomly divided into three groups and made models:C,normal control rats group;D,diabetic rats group;M,diabetic rats group treated with MMF,15mg/kg·d once daily by gavage. 24 hour urine protein excretion,blood glucose level and body weight were performed after 4 weeks of treatment.24 hour urine protein excretion, creatinine clearance ratio,blood glucose level and body weight were measured 8 weeks after induction of diabetes;Pathological changes were evaluated by HE stain in renal tissues;Immunohistochemistry was used to examine the expression of MMP-9 and TIMP-1.Result Renal tissues examined by HE stain and urine protein excretion of 24 hours,creatine clearance ratio,body weight were decreased in diabetic rats group treated with MMF as compared to diabetic rats group ,and the different was significant(P< 0. 05). Expression of TIMP-1 was significantly decreased(P<0. 01) , expression of MMP-9 was significantly increasd in the diabetic rats group treated with MMF(P< 0. 01).MMF also improved the histological changes of diabetic rats, prevent the fibrosis of renal .Conclusion MMF may help prevent the fibrosis progression of diabetic nephropathy,partially through regulating balance of MMP-9 and TIMP-1, up-regulating MMP-9 and down regulating TIMP-1. |
