Objectives:1)To extract and separate the effective component, used as a reference substance ;2)To determine the aldose reductase inhibition rate in each extraction from Salvia deserta Schang;3)To find an effective anti-diabetic complications part of Salvia deserta Schang;4)To set up the quality standard of effective part.Methods: 1)Extract the water-soluble parts from Salvia deserta Schang, separate the compounds by many kinds of column chromatography;2) NADPH in the 340 nm absorbance of the decline in value can be said that the activity of aldose reductase;3)Using the ultraviolet spectrophotometric(UV) method, take the total salvianolic acid as index ingredient, screen the effective part;4)Using salvianolic acid K as index ingredient and through high performance liquid chromatography(HPLC) made quantitative determination.Results:1) Obtained salvianolic acid K which purity is 98% from 80% MeOH extraction;2)80% MeOH extraction suppress to the aldose reductase rate is highest,the inhibition rate is 95.10% at 100μg/mL;3)The effective part is 10% EtOH-H2O part of 80% MeOH extraction which through the macroporous adsorptive resins column AB-8 and eluting by EtOH-H2O system;4)In the effective part, salvianolic acid K content is 638.98±5.56 mg/g through HPLC.Conclusions:1)salvianolic acid K comply with the request as a reference substance;2)Each extraction of Salvia deserta Schang and salvianolic acid K have shown good aldose reductase inhibition efficiency;3)The total salvianolic acid content>50%,conform to the traditional Chinese medicine crude drug fifth kind of new medicine standard;4) set up the quality standard for the first time,the method comply with the relevant regulations.
