ObjectiveTo establish a rabbit VX-2cancer animal model suitable for local treatment researchand study its biological characteristics.At the same time,to investigate the performance ofthe tumor models imaging and pathology,to find the research platform for the floor ofthe mouth cancer treatment.Methods(1) The establishment of tumor-bearing rabbits The establishment of tumor-bearingrabbits First of all, to recover the cryopreserved VX-2tumor tissue block, then toestablish the tumor-bearing rabbits to obtain tumor tissue blocks needed in the experiment.(2)The establishment of floor of mouth malignant animal models30New Zealandwhite rabbits, male or female rabbit age2to3months, weight1.5to2.0kg, were randomlydivided into2groups (n=15), were reared in standard laboratory;VX-2tumor tissue fromtumor-bearing rabbits who stripped, take and prepare a transplant of a certain size of thetumor tissue blocks and cell suspension, respectively, using the method of VX-2tumortissue implantation (group A) and the modified tumor cell suspension injection (group B)were inoculated in the middle of the district in the rabbit floor of the mouth to establish therabbit VX-2floor of the mouth carcinoma models.(3)outcome measures and statistical data To observe and to record the rabbits afterwaking hours, complications, rabbits spirit, appetite, activity, whether theabnormal;Respectively, after inoculation in rabbits one week, two weeks, three weeks, fourweeks to observe the general performance of the lesion,and to undergo the rabbit brain andneck CT andMR examination to observe with or without tumor formation, and to check thefloor of the mouth and neck with or without swelling and lymph node metastases, then,torecord the number,size, location of tumors found by the three means of naked eye, CT andMR scans,and to record the number,size, location of the transfer of lymph nodes found byCT and MR scans;To statistic tumor formation rate of the experimental rabbit;To statistic lymph node metastasis rate and lung metastasis rate of the experimental rabbit;To calculatethe rate of tumor growth,and compare the tumor rates and lymph node metastasis rate andlung metastasis rate of the two groups of rabbits; To observe the animals natural survivalsituation; To autopsy after death; Tumor, metastatic lymph nodes and organs by10%formalin-fixed and underwent pathological examination.Results26rabbits were found the lesion,assembly tumor rate of86.67%(26/30), Which thesuccess rate of the tissue block implantation (group A) was80.00%(12/15), the successrate of improved tumor cell suspension injection (group B) was93.33%(14/15), nosignificant difference in the two sets of vaccination success rate (P>0.05);In thisexperiment,26to20of the tumor rabbits were found neck lymph node metastasis, the totaltransfer rate was76.92%(20/26),10lung metastases, total lung metastasis rate was38.46%(10/26);Modified tumor cell suspension into the group of lymph node metastasiswas92.86%(13/14), lung metastasis was57.14%(8/14), are the highest; tumor tissueimplanted in the group of regional lymph node metastasis was58.33%(7/12), lungmetastasis rate was16.67%(2/12); Compared to tumor formation rate of the two groups ofrabbits, and compared to the cervical lymph node metastasis and lung metastasis of the twogroups of rabbits, were no significant difference. And the entire modeling process is simple,and inoculated with a high success rate. diet, mental activity and biological characteristicsof A and B two groups of tumor-bearing rabbit were no significant difference. Theobservation of the floor of mouth VX-2tumor animal models found that the VX-2tumorhas gone through three major phases: rapid tumor growth phase, the necrosis of the CancerCenter, surface ulceration stage,and the average survival period were about six to eightweeks.HE staining of biopsy of the tumor and lymph node specimens were observed and aredifferentiated squamous cell carcinoma, and confirmed into the tumor and lymph nodemetastasis.ConclusionThe rabbit VX-2floor of mouth cancer animal models by the establishment of the twoways of tissue block transplantation and improved cell suspension injection, and its rateof tumor, cervical lymph node metastasis, lung metastasis, growth rate, the nature of thetumor, tumor growth characteristics were no statistical significance. By CT and MRimaging means of the lesion dynamic detecting and evaluating the tumors, we canobserve of the occurrence and development of the lesion image changes more accurate and timely.The bottom of the rabbit mouth squamous cell carcinoma models established by theabove two operation methods,were easy to establish in a short time, and have stable andrepeatable biological nature, tumor formation rate, transfer rate, simple operation and manyother characteristics, its histological observation of performance for squamous cellcarcinoma,cancer biological characteristics of the population at the end are basically thesame, and other characteristics, provide large, good animal models for the experimentalstudy of cancer biology, therapeutics,and pharmacokinetics in the floor of the mouth.
