Objective:1.To explore the established method and identification of patient-derived xenograft models of the oral squamous cell carcinoma.2.Compare the efficacy of the HDST drug screening program and the PF chemotherapy program recommended by the NCCN guidelines for oral squamous cell carcinoma on the PDX model,and determine whether the recommended program of HDST technology is better than the PF chemotherapy program.Methods:After the surgery resection,the oral squamous cell carcinoma was treated with special treatment,and the inoculation needle transplantation was used to seed the immunodeficitamine mice,and the PDX model of export cavity squamous cell carcinoma was constructed and passed into the passage,and the P1-P3 was used for the deposition and identification.The consistency of tissue morphology and molecular phenotype of tumor tissue of PDX model were identified by HE staining and immunohistochemistry.The drug intervention experiment was carried out under the skin of the immunodeficiency mouse skin with a P4-P6 Generation Tumor,which was carried out to 100-200 mm3.Use the HDST drug sieve scheme to screen the most sensitive two kinds of chemotherapeutic drugs in each PDX model,and the PF chemotherapy regimen recommended by the NCCN guide is applied to the PDX model.The tumor length(L)and width(W)were periodically determined,and the moisture volume was calculated according to the formula,and the difference in tumor growth in each group was compared to the effects between the two programs.Results:The results of HE staining and immunohistochemistry of the tumor tissue of the P4 oral squamous cell carcinoma PDX model showed that the pathological and molecular phenotypic characteristics of the tumor tissue of the PDX model were basically the same as the original tumor tissue.After the drug intervention experiment,the three groups were compared.The tumor volume of the PF chemotherapy regimen group and the HDST chemotherapy regimen group was smaller than that of the control group,and the tumor volume of the HDST chemotherapy regimen group was smaller than that of the PF chemotherapy regimen group,and the measurement data of the two groups There is a significant difference between them(P<0.05).Conclusion:In the PDX model of oral squamous cell carcinoma,the tumor volume growth rate of the PF chemotherapy solution is greater than the HDST chemotherapy solution and the morphological characteristics are also more invasive,indicating that the chemotherapy solution given by HDST is better than the PF chemotherapy program.
