Objective:Through the investigation of the expression of stromal cell derived factor-1 and its receptor CXCR4 in the dermatomyositis lesion and in normal skin tissues,the role of the cytokines in the immunological disfuction of patients with dermatomyositis was explored.Methods:The expression of SDF-1 and CXCR4 were detected in the skin lesion of 22 patients with dermatomyositis and 15 nor- mal control by immunohistochemical technique (Avidin- Biotin -peroxidase Complex,ABC) and image analyse.Results:The SDF-1 and CXCR4 were presented in all specimens from the skin lesions and the normal controls. The expression of SDF-1 and CXCR4 in the dermal basal cell layer,stratum spionsum,granular layer,gland and the smooth muscle of the blood vessel of dermatomyositis lesion are significantly stronger,but low expressions are found in normal control.And the expression levels of SDF-1 in skin lesion were ( 0.2338±0.0707),which were greatly higher than those in normal contral (0.0861±0.0213), the expression levels of CXCR4 in skin lesion were(0.3630±0.0512), which were also markly higher than those in normal control(0.1192±0.0174).The expression have significant difference between dermatomyositis and normal control(both P<0.05).Conclusion:These findings indicate that up-regulate expression SDF-1 and its receptor CXCR4 might play an important role in the immunological disfunction of patients with dermatomyositis.These chemokine provide new targets for immune therapy.
