Objectives: To study the expression of CD133 in prostate tissues,the tissues of hyperplasia of prostate gland and prostatic carcinoma tissues。To.Find the relation between the expression of CD133 and clinicalpathology feature of prostatic carcinoma tissues and PSA。To Search the new tumor maker which can be used as the bases of early diagnosis and heal of prostatic carcinoma.Methods: 15 cases of normal prostate tissues ,21 cases of hyperplasia of prostate gland ,13 patients with A/B stage prostate cancer ,20 patients with C stage prostate cancer and 18 patients with D stage prostate cancer were collected. At the same time ,12 cases of prostatae well- differentiated adenocarcinoma,14 cases of prostatae moderately differentiated adenocarcinoma and 25 cases of prostatae poorly differentiated adenocarcinoma were also collected. By using immunohistochemical staining(Envision method) techniques, the expression of CD133 was detected and the relation between the expression of CD133 and clinicalpathology feature of prostatic carcinoma tissues and PSA were established.Results: all The basal cells and smooth muscle cells were dyed buffy 1 of 15 cases normal prostate gland cells was stained.Buffy.The positive rate is 6.7% .19 of 21 cases hyperplasia of prostate gland cells were stained buffy,The positive rate is 90.4% There was statistically significant difference between normal prostate gland and benign prostate specimens (P<0.01).13 of 13 cases cytoplasm of A/B grand prostate cancer cells were stained buffy and 3 cases nucelus of A/B grand prostate cancer cells were stained buffy 20 of 20 cases cytoplasm of C grand prostate cancer cells were stained buffy and 18 cases nucelus of C grand prostate cancer cells were stained buffy.18 of 18 cases cytoplasm of D grand prostate cancer cells were stained buffy and 17 cases nucelus of D grand prostate cancer cells were stained buffy.We find the expression prosition of CD133 antigen more in cytoplasm of A/B grand prostate cancer cells than in nucelus of that.The expression of CD133 antigen were transferred from cytoplasm to nucleus. 12 of 12 cases cytoplasm of prostatae well- differentiated adenocarcinoma were stained buffy and 3 cases nucelus were stained buffy. 14 of 14 cases cytoplasm of prostatae moderately differentiated adenocarcinoma were stained buffy and 12 cases nucelus were stained buffy. 25 of 25 cases cytoplasm of prostatae poorly differentiated adenocarcinoma were stained buffy and 25 cases nucelus were stained buffy. With the patho-grading decreasing ,the expression of CD133 were transferred from cytoplasm to nucleus. However, the level of PSA increased with the prostatic carcinoma patho-staging upgrading.Conclusion: 1. The sensitivity CD133 is better than PSA in normal prosate tissues and hyperplasia of prostate gland tissues.2. The expression of CD133 and PSA were positively correlated with clinical stage and the expression of CD133 was negatively correlated with histological grade in normal prostate tissues the tissues of hyperplasia of prostate gland and prostatic carcinoma tissues. CD133 may be used to assess the invasion of prostate cancer as a novel prognostic marker.
