Objective Investigate the expressions of CD133 and CD44 protein in the pericolonic tumor deposits and its clinical significance of colorectal cancer patients.Methods 90 colorectal cancer(CRC) patients who were treated in the first people’s Hospital of Chenzhou affiliated to University of South China from January 1, 2009 to December 31, 2010 were chosen as the research objects. All of the patients were divided into two groups, 60 cases diagnosed with TDs colorectal cancer and 30 cases without TDs. Collected the paraffin of cancer tissue, tumor deposit tissue, cancer adjacent tissues(tissue with in 3cm from the cancer tissue) and normal mucosa tissue(tissue with in 10 cm from the cancer tissue) of the 60 patients, and the cancer tissues of the patients without TDs. Used the HE staining method to diagnosed the tumor deposit, and immunohistochemical staining to detect the expression of CD44 and CD133 in these tissue samples. The clinicopathological data of this patients including gender, age, tumor location, histological type, the number of TDs, tumor differentiation, UICC staging, T staging, N staging, tumor size, Follow-up data. All of the statistical analyses were performed using SPSS 18.0 software. The clinicopathological of the patients’ were analyzed using chi-squared analysis. Kaplan-Meier estimate was used for calculated the survival rate of the patients and was compared between groups by Log-rank test. The independent prognostic factors of TD colorectal cancer patients were determined by multivariate analysis using the Cox proportional hazards model using stepwise selection to identify independent predictors. P<0.05 were considered significant.Results Both CD44 and CD133 protein were expressed on the cell membranes. CD133 was located on the cell membrane and the fragments of the tumor cells in the glandular cavity, Cytoplasmic expressions of cancer cells were very few. The CD44 cells were scattered or distributed focally along the side of the gland basement wall or the side of co-wall in CRC. There are no special features at the cells that expressed the protein CD133 and CD44. The groups of cancer tissue, tumor deposit tissue, cancer adjacent tissues have the high expression of CD133 and CD44 specimens, except the group of normal mucosa tissue. There is a significantly different at the expression of CD133 and CD44 between CRC patients with TDs or without TDs,while with no difference in the cancer adjacent tissues and normal mucosa tissue(P > 0.05). It’s no significant at the high expression rates of CD133 and CD44 between the groups of tumor deposit tissue and cancer tissues(P>0.05), while the high expression rates of CD133 and CD44 were obviously higher in tumor deposit tissue than in cancer adjacent tissues and normal mucosa tissue(P<0.05).The expression rates of CD133 were correlated to the N stage and UICC stage(P<0.05), while the expression rate of CD44 were correlated histological type, the number of TDs, UICC stage, N stage(P<0.05).There was no significant correlation between CD133, CD44 and sex, age, tumor location, tumor size(P>0.05). Data analysis show that Venerable age, histological type, the number of TDs T stage, N stage, UICC stage, CD133, CD44, CD133/CD44 were the prognostic factors of CRC with TDs. The prognostic factors with in univariate analysis were entered into the Cox proportional hazards model, the data analysis showed that CD133(P<0.05,HR=0.250, 95%CI=0.083-0.750), CD133/ CD44(P<0.05,HR=0.291, 95%CI=0.101-0.841) were the independent prognostic factors of CRC with tumor deposits.Conclusion 1. There is a significantly different at the expression of CD133 and CD44 between CRC patients’ cancer tissue that with TDs or without TDs, while with no difference in the cancer adjacent tissues and normal mucosa tissue. 2. There is no significant at the high expression rates of CD133 and CD44 between the groups of tumor deposit tissue and cancer tissues, while the high expression rates of CD133 and CD44 were obviously higher in tumor deposit tissue than in cancer adjacent tissues and normal mucosa tissue. 3. The expression rates of CD133 were correlated to the N stage and UICC stage, while the expression rate of CD44 were correlated histological type, the number of tumor deposit, UICC stage, N stage. 4. Univariate analysis showed that venerable age, histological type, the number of tumor deposit, T stage, N stage, UICC stage, CD133, CD44, CD133/CD44 were the prognostic factors of colon cancer with tumor deposits. 5. Cox regression analysis showed that CD133 and CD133/CD44 were the independent prognostic factors of CRC with tumor deposits.
