Clinical Observation On Cerebral Infarction And The Effect On Neurogenic Behavior And Nerve Cell Apoptosis With Cerebral Ischemia/Reperfusion Injury With Huoxue Recipe

In this paper, the clinical effects of Huexue Recipe in cerebralinfarction and further the effects on neurogenic behavior and nerve cell apoptosis withcerebral ischemic/reperfusion injury with Huoxue recipe were both appraised bymeans of clinical observations and experimental study.Methods: In clinical observations, the criteria of diagnosis and therapeuticeffects of disease and syndromes in traditional Chinese medicine, which have drawnup by SATCM and put up grade for cerebral infarction patients in the respect ofsyndromes and respects of nervous function. The 60 cases of patients enrolled wererandomly divided into 2 groups with prospective designing projects, the treatmentgroup was treated with Huoxue Recipe plus the basic treatment and the control onewith Aspirin proved by evidence medicine plus the basic treatment. Before and aftertreatment of Huoxue Recipe, the change of tongue demonstration, pulse condition,movements of extremity, function of language, blood-rheology and blood-lipid wereobserved. In order to guarantee security results or insecurity results about HuoxueRecipe, notes were taken about cardiogram and laboratory examination which wasmade up of blood routine examination, routine uronoscopy, renal function, liverfunction. The adverse reation was noted during the period of treatment.In experimental study, mice model of cerebral ischemia/reperfusion injury wasestablished by right common carotid artery blocking. The neurogenic behavior ofmice was observed, pathological changes of cortex and hippocampus were observedwith HE staining and light microscope, morphology of apoptosis nerve cell wasobserved by fluorescence microscope and percentage of apoptotic cell was measuredby TUNEL method; meanwhile, the quantitive assessment of apoptotic relative genebcl-2 and bax was made.Result and conclusion: The results of clinical observations showed that totalrate of clinical effects of Huoxue Recipe in cerebral infarction reached 90.8％, withthe single-blinded,random and controlled experiment. The clinical effects of HuoxueRecipe were shown as the four aspects below:①The sthenia syndrome of cerebralinfarction could be improved by Huoxue Recipe. The grade of Wind syndrome, Heatsyndrome, Phlegm syndrome, stagnation syndrome was cut down significantly(P＜0.01);②The neural function was improved and the grade of ADL was increased with Huoxue Recipe;③The content of blood-lipid, especially the content ofcholesterin in blood serum of cerebral infarction patients was reduced;④Thertheology of infarction patients was improved with Huoxue Recipe, which includedplasmic-viscosity, whole blood-viscosity and packed cell volume.The result of experimental study showed that the score of neurogenic behaviorof cerebral ischemic-reperfusio mice was higher than that of the sham operatedgroup(P＜0.05). Pathological changes such as cortical and hippocampal nerve cellregression, degeneration, and necrosis were found in the model mice, percentageincrease of hippocampal nerve cell reached the peak after 6 hours of the operation.The experiment showed, Huoxue recipe could effectively improve the neurogenicfunction, lower the percentage of apoptosis significantly compared with those in thecerebral ischemic-reperfusion group, Bcl-2 positive protein cells were increasingwhile Bax positive protein cells declining significantly compared with the the cerebralischemic-reperfusion group(P＜0.01). Huoxue recipe had the effect of improving theneurogenic behavior, reducing the cell apoptosis rate of cortex and hippocampus inmice with ischemia-reperfusion injury.
