ObjectiveCervical intraepithelial neoplasia is the precursor lesions of cervical cancer. It isthe main way to prevent cervical cancer by early diagnosing and effectively treatingCIN.The way to diagnose CIN includes clinical test, cytologic test, biopsy bycolposcopy, pathology diagnosis and necessary PCR-DNA test. With the mature ofdiagnosing CIN, cytologic test combining biopsy by colposcopy is still credible methodto early diagnose CIN and cervical cancer A lot of research in home and abroad indicatethat high-risk research is the direct cause of CIN and cervical cancer.The research is confirmed by pathologist DR CJLM meijer in Holland.The negative predictive value of diagnosis of CIN by high-risk HPV test is morethan 99%. The rate of screening CIN by detecting high-risk HPV combining cytologictest is higher than that only cytologic test. It indicates that the rate of screening. CIN bytwo me thuds is nearly 100%.Methods1. SubjectivesThe clients came from outpatient of Liaoning Cancer hospital from June 2005 toDecember 2005.With their cervical disease confirmed by colposcopy. Totally 93 cases. CINⅠis 31 cases, CINⅡis 31cases, CINⅢis 31 cases.2. Test HPV DNA with hybrid capture methodHybrid captureⅡHPV DNA system and test box for high-risk HPV are suppliedby Digene corporation. Probe comprises of 13 subtypes, type 16, 18, 31, 33, 35,39, 45, 51, 52, 56, 58. The main steps are (1)spallation(2)hybridisation(3)capture(4)response(5)flush(6)collecting sign(7)outcome judgment: the content of HPV DNA isabove 1pg/ml, which means the ratio between RLU and CO is above 1, positive.3. Thinprep cytologic testCells from cervix is collected with the special brush, the specimens are conservedin a bottle with preservative fluid. After making thin-laver, being fixed by 95% alcOhol,pap staining, diagnosing by pathophysiological doctors, the specimen classified withASCUS and above are positive.4. Pathology by colposcopyBishops from aceto-white epithelium, mosaic, white glade, atypical vessels and 4points of transformation zone from normal women are sent to check for the lastdiagnosis.5. Stastical analysisX~2 test is applied(α=0.05) and software SESS 12 is used for data analysis.Outcome1. The ratio of high-risk HPV detected from CINⅠgroup is 51.6%, CINⅡis58.1%, CINⅢgroup is 93.5% in conclusion. infection of high risk HPV is relative tooccurrence of CIN and disease are wore serious infection rate of HPV is higher.2. In the group infected by HPV. Positive ratio of cytologic test is 58.3% inASCUS. 72.4% in LSIL. 95% in HSIL, there is a significant difference between groupinfected by high-risk HPV and group not infected by HPV (P<0.05) Conclusion1. Infection rate of high-risk HPV is ascending with serious degree of cervicaldisease, and the difference is significant.2. With the grade of cytology increasing, the positive rate of high-risk is higher.3. Sensitivity of diagnosing CIN by HPV test is high, positive ratio of screeningCIN can be improved by detecting high-risk HPV with cytologic test.
