Objective: Analyzing the two-dimensional gel electrophoresis (2D-PAGE) with cerebrospinal fluid(CSF) of Parkinson's Disease patients (treatmenting with and without drugs) and controls (patients with headache). Selecting significant differentially displayed protein spots and identifiying by MALDI-TOF-MS. Analysis the disease-specific proteins(DSPs).Methods: proteins extracted from CSF was focused immobilized pH gradient isoelectic focusing (IPG-IEF) as the first dimension, and then was electrophoresised in vertical flat sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) as the second dimension. The protein spots in gels was visualized by silver staining gel followed by gel scanning and image analysis. Searching for differentially displayed protein spots then identifiying by MALDI-TOF-MS. Analyzing DSPs roles and effect.Results: Satisfied 2D-PAGE patterns of protein in CSF with PD patients and controls was acquired in the experiment. Using Pdquest software analyze the 2D-PAGE and findout three DSPs. They are transthyretin, apolipoprotein E and serotransferrin.Conclusion: There are some significant difference protein spots in 2D-PAGE gels with Parkinson's Disease and controls. Durgs have little effective to protein patterns in cerebrospinal fluid.
