Objective:①to collect the cerebrospinal fluid of Parkinson's disease patients, non-Parkinson's disease patients, and control group.②Then to produce electrophoretograms;③to select the different protein points in the gels and to explore the diagnostic biomarker.Methods: Firstly, to enlist 54 people randomly. Then divide them into 5 groups. The Parkinson's disease group comprises 27 Parkinson'disease patients. The non-Parkinson's disease group is composed of 18 patients M+SE=53±2.2 y , male/female:10/8. The control group is composed of 9 patients(M+SE=50±4.25 y,male/female:6/3). This experiment applies the improved Webster scale and Hoehn &Yahr scale to evaluate the PD patients and divide the patients to 3 smaller groups: mind group (14 patients, M+SE=56±2.6y,male/female:9/5), moderate group (8 patients, M+SE=57±4.7y,male/female:=5/3), and severe group (5 patients, M+SE=59±5.2y,male/female =4/1), The non-Parkinson's disease group consists of 3 patients suffer from Restless legs Syndrome, 3patients suffer from Wilson's Disease, 3 patients suffer from Congenital cerebella ataxia, 3 patients suffer from Parkinsonism, 3 patients suffer from essential tremor, 3 patients suffer from infarction of the brain. The control group consists 9 patients suffer from tension headache. This experiment utilizes two-dimensional polyacrylamide gel technology that bases on the immobilized pH gradient and sodium dodecyl sulphate to separate the cerebrospinal fluid of each group. Then dye the gels by silver staining and analyze the gels by PDquest8.0. Finally identify the different protein points by MALDI-TOF-MS and explore the function of these protein points.Results: The experiment finds 789±32 points in gels of PD groups. 4 protein points have conspicuous discrepancy between the Parkinson's disease group and non-Parkinson's disease group, control group, the emergency rate of the 4 proteins is 100%; 3 protein points decrease and 1 protein increases. The Mascot database indicates the descending protein points are DNA-directed RNA polymerase III subunit RPC5(RPC5);Dual serine/threonine and tyrosine protein kinase(DUSTY) (P < 0.05), Activity-regulated cytoskeleton-associated protein(ARC,Arg3.1) (P<0.05), the ascending protein point is G2/mitotic-specific cyclin-B1(CCNB1) (P<0.05).Conclusion:①The protein points in CSF of Parkinson's disease patients are well separated by immobilized pH gradient gel in PH 4-7.②The protein points has conspicuous discrepancy between mild group ,moderate group and severe group;③Compared with non-Parkinson's disease group and control group, it is indicated that there are 4 different protein points exsit in each group of PD, it is identified that those proteins are functional in protein transcription, the plasticity of nerve cell as well as the risk suffering from depression and the regulation of cell cycle.④the 4 different protein points exists in the whole disease course of PD and they may be the diagnostic biomarker of PD that distinguish the PD patients from other neural diseases and normal people with sound repeatability and stability.
