Effect Of Xinnaojin Injection On Expression Of NOS1 In Cerebrum And The MRNA Expression Of B-Amyloid Precursor Protein In The Hippocampus In Rats With Alzheimer's Disease

Objects:To observe the effect of xingnaojing parenteral solution on the behavior, the expression of NOS1 in cerebrum, and the mRNA expression ofβ-amyloid precursor protein (APP) in the hippocampus in rat model with Alzheimer's disease (AD).Method:42 rats were divided randomly into 4 groups: blank control group(10), model group(12),Xin Naojing low-dosage group(10) and Xin Naojing high-dosage group(10). Model group, low-dosage group and high-dosage group was injected in peritoneum with D-galactose(60 mg·kg - 1·d– 1, prepared with sodium chloride)and was administrated with aluminum chloride(100 mg·kg - 1·d - 1, prepared with double distilled water)in 90 days; Control group was injected in peritoneum with equal volume D-galactose and was administrated with equal volume aluminum chloride. In fifth week, Xin Naojing low-dosage group and high-dosage group was injected respectively in caudal vein of rats with Xin Naojing 1 ml·kg - 1·d– 1 and 2 ml·kg - 1·d– 1 for 8 weeks; control group and model group was injected with equal volume Sodium Chloride. Every group was administrated in the morning and the experiment last for 12 weeks. After 12 weeks, Morris water maze was used to measure the behavior of rats, histochemical technique was used to observe the expression of neuronal nitric oxide synthas (NOS1) in brain, and RT-PCR was used to assay the level of APP mRNA in the model rat hippocampus.Result:In Morris water maze, escape lantency of medication administration team(MAT)(25.6±14.8,21.5±15.1)is notably decurtated,compared with model group(31.7±29.3)(P <0.05);In spatial probe ,the frequency that the rats go through the place where the platform was placed before is increased in MAT(6.8±2.7,8.6±1.3) compared with model group(4.7±1.8)(P <0.01); The time that the rats in MAT(40.55±18.13,45.74±9.82) stayed the quadrant where the platform was placed before has not significant deviation with that in model group ( 26.65±13.28 )(P>0.05);Compared with model group(cortex:4.34±1.71, hippocampus: 3.98±2.15),Xin Naojing can strengthen NOS1 activity of center cortex(7.76±2.68,8.43±1.16) and hippocampus(6.52±1.36,9.67±1.49) ,and reinforce density of neuron(P <0.05-0.01); The expression of APPmRNA in hippocampus in MAT(1.108±0.029, 0.986±0.023) is lower than that in model group(1.359±0.035)( P <0. 05). Conclusion:The activity of NOS1 in central nervous system and the expression of APPmRNA in hippocampus has significantly close correlation with the ability of learning and memory;Xin Naojing could improve the disorder of learning and memory in mice with spontaneous senile dementia significantly. Xin Naojing could enhance the activity and neuron density of NOS1 in the central cerebral cortex and hippocampus, and all these efects had a dose-effect relationship. The APPmRNA expression level was significantly lower in low-dosage group and high-dosage group compared with model group;Xin Naojing parenteral solution can improve the disorder of learning and memory in mice model with spontaneous senile dementia significantly.
