In order to further study on the essence of hanxie and its correlated factors invading lung,we carry on an experiment of simulating hanxie,which changes with different temperature gradient changes and its correlated factors-fengxie and shixie to stimulate healthy rats in the laboratory.In the experiment,224 rats were randomly divided into 28 groups,each 8:normal temperature group,stationary low temperature groups(LT groups),sudden decline of temperature groups(DT groups),slow decline of temperature group,LT with fengxie groups,LT with shixie groups,LT with fengxie and shixie groups,DT with fengxie groups,DT with shixie groups,DT with fengxie and shixie groups.All LT groups were divided into 3 gradient groups:10℃,0℃,-10℃again.All DT groups were divided into 3 gradient groups:20℃~10℃,20℃~0℃,20℃~-10℃again.Put each group of rats in the thermostat corresponding condition to stimulate.4 days later,gather the specimen. Observe the rats' behaviour,the pathomorphological changes of lung and content of IL-6,TNF-α,IL-4,IFN-γin lung homogenate.Fractalkine mRNA in lung homogenate was detected by reverse transcription-polymerase chain reaction (RT-PCR).Expression of fractalkine protein was observed by immunohistochemistry. Experimental datum are analyzed by SPSS11.5.The results manifest that hanxie not only have something to do with stationary low temperature,but also with decline of temperature closely.Along with temperature lower and difference in temperature larger,hanxie do harm to lung more seriously.And sudden decline of temperature do harm to lung more seriously than slow decline of temperature.A certain fengxie and shixie can aggravate the injury of lung tissue caused by hanxie.Using the methods of immunohistochemistry and RT-PCR,we prove that fractalkine,which is a member of chemokines,has a low expression in the lung tissue of normal temperature group rats.The level of fractalkine mRNA and expression of fractalkine protein in the injured lung tissue of each model group increased along with lung injury aggravating. Hence we conclude that fractalkine performs a vitally important role in lung injury.
