Objective: In an attempt to separate the serum lipoprotein and their subfractions with each particle as a whole, the balance among which could be quantitated and analyzed, and to investigate the relationship between the relative amount of each Lp subfraction and acute myocardial infarction(AMI). Methods: To separate the serum lipoproteins prestained with Sudan Black B in the first double gradient polyacrylaniide gel electrophoresis (f- DGPAGE). There were 11 fractions and subfraction containing nonesterified acid albumin(AL), alpha-lipoprotein 1 ( a Lp1 ), P Lp, inter-beta- lipoprotein(int P Lp), pre-betalipoprotein l-2(pre P Lp,-2) and Chylomicrons (CM). The electrophoretic zoi~es on polyacylaniide gels were measured densitometrically and it's images were acquired by the gel acquisition system .The densitometric quantitation of Lp aubfractions had been finished. In 36 cases with AMI conformed to the WHO diagnostic standard , 21 cases with old myocardial infarction, 7 cases without CHD, 16 normal controls, the serum concentrations of triglyceride(TG), total cholesterol(TC), high density lipoprotein cholesterol(I4DLc) and low density lipoprotein cholesterol (LDLc) were measured or calculated after an fast. The first serum lipoprotein electrophoretagrani(f-SLpG) were synchronously measured by the f-DGPAGE. Results: (1) The f-DGPAGE could separate the strum lipoproteins into about 11 zones. The relative amount of Lp and it's subfrction in SLPG could be quantitated .(2) Compared with that in normal cont o1 gro .lp and(or) non- CHD group, the AL were significantly higher in 44M1 group, and the sequence of a Lp1..5 disordered(P< 0. 00 1. 05). so as that in the AMI subgroup without routine dyslipidemia. Conclusions: (1} The f-IXIPAGE can separate the Lp into 1 1 fractions named AL, a Lp15, P Lp, mt P Lp, pre P Lpj-21nd ideally show its heterogeneity. (2) The AL were significantly higher in1 AMI group. The AL is actually the complex of FFA and albumin, and the hight of FFA could be the secondly reaction of AMI. (3) a Lp maybe the nqonate HDL. Both the heighter and the lower could be a abnormal state. (4). The irxreased relative amounts of LP in Lp electrophoretagram and SLPG are significantly correlated with AMI. (5) the amount of disordered sequence of a Lp15 were 33.3% in all AMI , which show the quantitative balance among Lp were in disequilibrium state. (6) Lp electrophoretagram may p a new experimental index and used for the clinical diagnosis of dyslipicfefriia and syndrome ofintraspleen ashtenia(ISA) in traditional Chinese medicine.
