Objective:To difme whether three kinds of purified bentonite can be used as direct compression excipients,through evaluating the size distribution,aerated density,tapped density,Repose angle,and compactibility of three kinds of purified bentonite;To study the water uptake characters of three different kinds of purified bentonite and some other disintegrants and mixed disintegrants;To investigate the disintegrability of three kinds of purified bentonite used as filler and disintegrants when occupied different percentage in the calcium carbonate tablets;To test the properties of purified bentonite as the disintegrants of dispersion tablet by Investigating disintegrate time and the dissolution rate.Methods:To test the compressing process of purified bentonite by Kawakita formula,The tablets of purified bentonite were prepared by direct compression.The hardness and compactibility property of the tablets were evaluated;The Measurement of water uptake by bentonite or some other disintegrants and mixed disintegrants was designed according to a reference.Meanwhile,study their swelling properties;the calcium carbonate tablets that three kinds of purified bentonite used as filler and disintegrants occupied different percentage were compressed at different pressure,The hardness and disintegration time of the calcium carbonate tablets were tested respectively; Mixing bentonite,MCC,L-HPC,PVPP and CMS-Na respectively in different proportion, choice the mixed disintegrants that has good water uptake characters(which has the high degree that mixed disintegrants was saturated by water within 3 min)as the disintegrants of ibuprofen dispersible tablets,Using the uniform design for selecting the best amounts of the mixed disintegrants and the amount of other addition,the dissolution rate, suspensibility and disintegration time were as index.Results:The flow ability of three kinds of purified bentonite was poor than some other commonly used excipients of tablet. The particle size of A were smallest.C was the second,aerated density of three kinds of purified bentonite was lower than Veegnm but tapped density were higher than Veegum. The compactibility of three kinds of purified bentonite and Veegum were worse than MCC and PCS;Three kinds of different purified bentonite,Veegum,CMS-Na and corn starch have the similar water uptake process,it's slow to be saturated.By contraries,L-HPC, PVPP are saturated within 4 seconds.MCC is a special one which is different with both of them;without improve tabletting pressure,The hardness of the calcium carbonate tablets with three kinds of purified bentonite occupied 5%in the tablets were higher than the tablets compressed with some other fillers or without filler.(P<0.01),The dissolution of the ibuprofen dispersible tablets was fast,could,be completely form into suspension, which could Screen through,the 710um mesh.Conclusion:Three kinds of purified bentonite can not be used as direct compression excipient;three kinds of different purified bentonite have the similar water uptake performance than Veegurn or better.Three kinds of purified bentonite can be used as dry binder and disintegrants in tablet,A is better than B and C;purified bentonite A could be used as the disintegrants of dispersible tablet.
