Objective::to study the correlation between different extract manner and effect of pharmacology of xinjiang Vernonia anthelmintica(VA).Method:(1)the samples were prepared by solvent extraction,we got three extracts:water extract(Q1),acetone extract(Q2)and residual material(Q3).(2)B16 murine melanoma cell was cultured in vitro,MTT was used to study the proliferation,the tyrosinase activity was detected by Oxidative DOPA reaction,the melanin content was measured by NaOH method.(3)Then we choosed C57BL mouse as the experimental model to observe the active fraction of VA on the skin and hair follicle.(4)the mechanism action was further studied in the enzymology and western blotting.(5)the material basic of effect was analyzed by HPLC. Result:(1)Q1 and Q2 could significantly increase cell proliferation,while the effect of Q3 is slightly.The tyrosinase activity tests showed that Q1 and Q3 stimulated the activityand increased melanin content,20~80μg,ml-1Q3 would obviously increase melanin content(P<0.01,comparable with control grope).(2)we observed that Q3 in the concentraction of 48.17mg.kg-1,96.34 mg.kg-1,192.68 mg.kg-1can enhance the synthesize of melanin granules in hair follicle of C57BL mouse,(3)western blotting result showed that Q3 can enhance the expression of tyrosinase protein.(4)It was duduced that the effect of proliferation is relate with the peak of 1,2,3,4,5,11,16, 17,19,20,21,23,25,26,while the tyrosinase activity and melanogenesis is correlated with the peak of 12,14.Conclusion:A relationship between different extraction methods and bioactivity of melanogenesis was showed,while Q3 can significantly enhance melanogenesis in vitro and vivo,it was initially deduced that the mechanism of its effect was via up regulate activity of tyrosinase and expression of tyrosinase protein.It was showed that the manner,effect of pharmacology screening binding with drug extract, was important to reveal the biological active fraction in developing natural medicine.
