| Objective: Avascularity is the key feature of the cornea. Corneal neovascularization would develop because of infection, surgical trauma,etal. Acuity of vision would be seriously damaged. The mechanism of neovascularization is multiplicity. Wilderness growth factors and regulatory factors participate this process, and keep balance between enhancement and containment. Matrimetalloprdeinase-2,9 are demonstrated to be important in the process of CNV by some researches recently. Doxycycline is the most active inhibitor of MMPs applied to clinic at present. This experiment is considering the effect of the doxycycline for alkali burn of the rat cornea by using eyedrop application of different concentration gradient of doxycycline to investigate the effective and approach the mechanism of action.Methods: 67 Wistar rats were selected for experiment: 5 were common contrast, 62 alkali burn of the rat cornea model was established. 32 were divided into four groups at random,. Four groups were droped 0.1%,0.5%,and 1% doxycycline humor ocular to the three treatment groups' cornea and saline to the control group's. The control group were droped PBS humor ocular.30 were divided into RT-PCR1 group and RT-PCR2 group,RT-PCR1 were droped 0.5% doxycycline humor ocular, RT-PCR2 were droped PBS humor ocular. All groups have been droped one gutta 3 times per day for 28days.All groups have been observed the corneal opacity and cnv by slit-lamp everyday for 28days.The area of NV and the opacity of cornea would be analysised by statistics selecting the corneas at day 4,7,14,21,28 after corneal alkali burns.2 groups were killed at day 2,7,14.The expression of MMP-2,9 mRNA were examined by reverse transcription-polymerase chain reaction(RT-PCR). To compare the differences of 4 groups using analysis of the data. Results: 0.5%,1% doxycycline eyedrops could inhibit the corneal neovascularization and opacinity of cornea. The analysis of thedata of RT-PCR demonstrate that the expression of MMP-2,9 mRNA were different between the 0.5% treatment groups and the control group.Conclusions: 1. Doxycycline eyedrops can inhibit the formation and growth of CNV.2. Doxycycline inhibits CNV referring to inhibit the expression of MMP-2,9 mRNA. |
