Corneal neovascularization (CNV) is a severe complication induced by various pathogenetic factors. This condition is associated with severe visual impairment and is a high-risk factor in rejection after allograft corneal transplantation. Therefore, it has always been a major field to study the mechanisms of corneal neovascularization and the effective way of treatment.Partâ… Inhibitory effect of ginsenoside Rg3 on rat corneal neovascularizationObjective To evaluate the antiangiogenesis of ginsenoside Rg3 in rat corneal neovascularizaiton.Methods In Sprague-Dawley rats, inflammatory corneal neovascularization was induced by cautery with alkali. Animals were randomly divided into 4 groups, the gensenside Rg3 (Rg3) group (0.1%, 0.2%, 0.5%) and the control group. The area of CNV detected by slit lamp microscope in rat cornea at the different times(at 1, 4, 7, 14d after cautery). Then the rats were sacrificed at the different day and the corneas were taken for pathological analysis.Results The growth of CNV can be inhibited significantly in different Rg3 groups. The Rg3(0.5%) is the most effective one in treatment groups. (1) Compared to the control group, the area of CNV was significantly reduced with Rg3 (0.5%) on 1, 4, 7, 14 day (P<0.05). (2) The pathological analysis shows that the amount of inflammation cells and newly formed vascular in the rat cornea of the experimental group (0.5% Rg3) are lower than that of the control group.Conclusion Topical application of ginsenoside Rg3 has a significant antiangiogenic effect against CNV induced by alkali burn. Animals were randomly divided into 3 groups (the normal group, the control group and the treatment group). The rats were sacrificed and the cornea were taken on 1,4, 7, 14 day. Immnuohistochemistry method and relative quantative reverse transcription polymerase chain reaction were used to investigate the distribution and expression of VEGF and Flk-1 in the rat cornea at the different times.Results The cornea of normal rats expressed slightly or did not expressed VEGF and Flk-1. In the control group, The eptithelium, stroma, endothelium and the endothelium cells of neovascularization of cornea showed staining of VEGF and Flk-1 by immnuohistochemistry. The expression of VEGF mRNA and Flk-1 mRNA have correlation at the different times.Conclusion Ginsenoside Rg3 inhibit the rat corneal neovascularization effectively by downregulating the expression of VEGF and its receptor Flk-1.
