Experimental Research On Different Solutions Used In Preservation Of Amputated Limbs

Objective:to study the difference between UW(University of Wisconsin) solution and the self-prepared perfusing-fluid in amputated extremities perfusion, and if it can substitute the expensive UW solution,to be economically used in clinical application.Methods:selecting 24 healthy adult rats(sex is ignored,250-300g Weight), provided by GuangXi Medical University Laboratory Animal Center,to be randomly divided into 4 groups,every group consist of 6 rats.First group;as a control sample,without any solution processing,the second group;6 rats amputated posterior extremities of 6 rats,were perfused with "Isotonic sodium chloride solution mixed with Heparin",the third group;6 rats amputated posterior extremities were perfused with "Ringer solution mixed with heparin and dexamthasone",fourth group;amputated posterior extremities of 6 rats,were perfused with UW solution.After the amputed extremities were perfused,they were refrigerated in 4℃,dry,and avoiding any direct contacting with ice to avoid harming the viability of the cells.Then by examining the samples to detect any ischemic changes,using the light microscope to differenciate any general structural changes also using the electronc microscope to detect any ultrastructural changes,as well as to observe any biochemical indicator changes, then finally using SPSS13.0 program for statistical processing.Results:Using self prepared solution to keep the viability of amputated extremity,no matter using chemical standard or under light /electronic microscope to detect the structural changes,going through statistical processing, we find out that the perfused amputated extremity groups were better than the control groups;between the three perfuse-groups there were no variability found out.Conclusion:the groups of perfused-extremeties were better than that not perfused(control sample).The self-prepared solutions and the UW solution results were to great extent equivalent,and therefore can be used in clinical practice as a replacement choice.
