Background and purpose: nasopharyngeal carcinoma(NPC) is the most common malignant tumor in our country. With the development of oncomolecularbiology, we gradually discover that occurrence mechanism of nasopharyngeal carcinoma is affected by hereditary predisposing genes polymorphism and environment factor at the same time. Recently the significance of VEGF gene and Survivin gene are paid close attention by most researchers as they both are considered as the tumor associated gene, which many studies show that both genes have relationship with occurrence and development of lots of tumors. The objective of this study is design to investigate the expression of VEGF protein and Survivin protein in nasopharyngeal carcinoma and nasopharyngeal mucosa tissue of chronic inflammation by the way of immunohistochemistry staining, in order to approach the two proteins' effect and significance of the occurrence and development in nasopharyngeal carcinoma and hope to give theoretical guidance to the diagnosis, therapy and prognosis in the clinic practice of nasopharyngeal carcinoma.Material and method: In this study, 105 cases of paraffin blocks of nasopharyngeal carcinoma were selected from the Department of Radiotherapy in Guangxi Medical University affiliated tumor hospital in the period of years from 1998 to 2000. All the patients had been confirmed through pathological examination and received radical radiotherapy first. At the end of radiotherapy, they were confirmed there were no tumor tissue remained in the pharynges by the macroscopic examination. Followed up from 5 to 7 years , the rate of following up is by 95%. We are going to use immunohistochemistry staining to check the expression of VEGF protein and Survivin protein. The methods that are used to analysis the data are chi-squre test, correlation analysis, Cox regression, survival analysis. The analysis is done by SPSS version 13.0 software.Results:(1) The positive expression of VEGF and Survivin protein in nasopharyngeal carcinoma are 76%, 58%, respectively, which have statistical significance between carcinoma group and normal tissue group.(2)Intensity of the expression of VEGF protein is relate to lymph nodes metabasis, tumor stage, cranial nerve palsy, basalis encroachment (P<0.05) and not relate to gender, age, encroachment in front or back of the belemnoid, VCA-IgA. The expression of Survivin protein has no significant correlation with gender, age, lymph nodes metabasis, tumor stage, cranial nerve palsy, basalis encroachment, encroachment in front or back of the belemnoid, VCA-IgA(P<0.05). (3)The negative expression of VEGF protein or Survivin protein has better 5 years survival rate than positive expression of it , low expression of it has better 5 years survival rate than high expression in clinic, however the survival function have no evident significance in statistics among the groups(P>0.05).The negative expression of VEGF and Survivin proteins at the same time has better 5 years survival rate than high positive expression of them, there is statistical significance between survival curve in two groups(P<0.05).(4)Cox regression indicate that the expression of VEGF protein or Survivin protein is not evidently associated with the survival of the patients with nasopharyngeal carcinoma (P>0.05). (5)There are a significant correlation between the expression of VEGF and Survivin (P<0.05).ConcIusion:(1)The abnormal expression of VEGF and Survival play a certain role in course of the occurrence and development of nasopharyngeal carcinoma.(2)The study with more cases and longer time observing is necessary for the effect of prognosis coming from VEGF and Survivin gene in nasopharyngeal carcinoma patients. (3)There may be some certain inter-regulation mechanism between VEGF and Survivin genes.(4)Investigation of the expression of VEGF protein and Survivin protein at the same time maybe is a good way estimating the prognosis in NPC patients.
