Expression And Correlation Of Survivin,Caspase-3 In Nasopharyngeal Carcinoma

Objective:To investigate the expression of Survivin and cysteine protease-3(Caspase-3) in NPC(nasopharygeal carcinoma) and chronic nasopharyngitis tissues by immunohistochemical method(ELPS method).Aiming to explore the role of Survivin and Caspase-3 in the pathogenesis of NPC and provide scientific basis for the early diagnosis and prognosis of NPC(nasopharygeal carcinoma).Methods:30 cases of NPC removed by video-naso-pharynogo-laryngoscopes biopsy in otolaryngology of the affiliated hospital of Guizhou medical university from September 1,2013 to December 31,2013 were regarded as the experimental group.Each specimen was diagnosed by Pathology Department of the affiliated hospital of Guizhou medical university,11 cases of female,19 cases of male,15-72 years old(average age 47 years old).All preoperative patients were not carried out preoperative long-term oral non-steroidal anti-inflammatory therapy,chemotherapy and biological target therapy.30 cases of suspected NPC patients removed by video-naso-pharynogo-laryngoscopes biopsy,which were diagnosed as chronic nasopharyngitis by department of pathology,were regarded as control group.The method of immunohistochemical method(ELSP) was used to test the expression of Survivin and Caspase-3 in NPC and chronic nasopharyngitis tissues.Results:1,In the NPC tissue,the positive expression rate of Survivin was 80.00%(24/30).In the chronic nasopharyngitis tissue,the positive expression rate was 26.67%(8/30).The difference was statistically significant(Z=-4.75,P<0.01).The positive expression of Survivin in III-IV level clinical stage of nasopharyngeal carcinoma tissue was higher than it in I-II level clinical stage of nasopharyngeal carcinoma tissue.(P<0.05)The positive expression of Survivin in Lymph node metastasis tissue was significantly higher than it in the tissue without lymph node metastasis.(P<0.05).The positive expression of Survivin in distant metastasis tissue was significantly higher than it in the tissue without distant metastasis.(P<0.05)The expression of Survivin in nasopharyngeal carcinoma had no significant correlation with sex,age,tumor location(P>0.05).2,In the NPC tissue,the positive expression rate of Caspase-3 was 30.00%.In the chronic nasopharyngitis tissue,the positive expression rate was 93.33%.The difference was statistically significant(Z=-5.28,P<0.01).The positive expression of Caspase-3 in lymph node metastasis tissue was significantly lower than it in the tissue without lymph node metastasis.The expression of Caspase-3 in nasopharyngeal carcinoma had no significant correlation with sex,age,tumor location,distant metastasis and clinical stage(P>0.05).3,There was negative correlation between expression of Survivin and Caspase-3 in nasopharyngeal carcinoma(r=-0.378,P < 0.05).Conclusion:1,The expression of Survivin and Caspase-3 may play certain roles in the initiation and development of nasopharyngeal carcinoma.2, By the Combined detection of the expression of Survivin and Caspase-3 may be valuable for diagnosis and prognosis nasopharyngeal carcinoma.
