Objective To explore the cell clonal origin of multinodular hepatocellular carcinoma and assess its values and clinical significance using PCR-SSCP to detect the expressions of TP53(exon6,7),bcl-10,E-adherin,β-catenin genes among the nodules.Methods PCR-SSCP was used to detect the expressions of TP53(exon6,7),bd-10,E-cadherin,β-catenin genes in various nodules in 25 patients with multinodular hepatocellular carcinomas(experimental group,EG),two pieces of tumor tissues in 25 patients with single nodule hepatocellular carcinoma(control group,CG).Nomal hepatic tissues in 15 healthy people respectively(nomal group,NG),statistical ananlysis was performed using SPSS 13.0.Results The mutation rate of TP53 exon7,bcl-10 genes had no differences between EG and CG(P>0.05);TP53(exon6),E-cadherin,β-catenin genes had low mutation rate;In EG,16 cases were diagnosed metachronously multicentric occurrence(MO)and 9 cases were intrahepatic metastasis(IM)by multigenic detection;There was a higher rate of complicated lesions with chronic hepatitis and cirrhosis and a lower recurrence rate than those in CG(P<0.05).Conclusions The detection of TP53 exon7,bcl-10 genes using PCR-SSCP combined clinical data can identify the cell clonal origins in multinodular hepatocellular carcinomas.There are different cell clonal origins exist in multinodullar hepatocellular carcinoma,the MO has a better therapeutic effect and prognosis than that in IM.Combined with clinical datas,PCR-SSCP is useful for the selections of therapy and assessments of prognosis.
