The major concern in life scientific research inevitably changes direction from gene to protein since the accomplishing of Human Genome Project. The conventional manner of investigating one single protein is not adequate to the requirement of modern post-genome era. It is necessary to investigate proteins in levels of entirety, dynamics and network. The newly emerging proteomics was developed on the basis of two-dimensional electrophoresis, by which the cellular protein mixture was separated according to their different isoelectric points and molecular weights. Through the Mass-spectrometry technique and bioinformatics, the unknown proteins were identified. With the application of proteomics techniques in the research of neoplasm, some differential expression proteins would be identified by comparative analysis of the proteome between normal and neoplastic tissue, and some tumor-specific markers and therapeutic target may be picked out by further analysis of these proteins, it will be helpful to clarify the mechanism of tumorigenesis.This study is a primary proteomics investigation of papillary thyroid carcinoma(PTC). Protein extracts of papillary thyroid carcinoma and normal thyroid tissues from 10 patients were separated and identified by proteomics technique. With comparative analysis of 2D-PAGE gel images between PTC and NT tissues by using ImageMaster software, the basic 2-DE images of PTC and NT was obtained, and a considerable difference of quantity and distribution was found from the basic 2-DE images of PTC and NT. Four potential differential expression spots were selected for Mass-spectrometry identification. Among them, Annexin I, Peroxiredoxin 1, Mitochondrial aconitase were up-regulated while Carbonic Anhydrase I was down-regulated.
