Objectives : The differentially expressed genes in papillary thyroid carcinoma and corresponding paracancerous tissues were screened by gene chip,and the expression of the genes with the largest difference multiple in papillary thyroid carcinoma was analyzed by bioinformatics method.Finally,the expression of papillary thyroid carcinoma and its corresponding paracancerous tissues were further verified by immunohistochemical method,the pathogenesis of papillary thyroid carcinoma was preliminarily clarified,and the genes related to the occurrence of papillary thyroid carcinoma were explored.It provides theoretical basis and reference for improving the therapeutic effect and prognosis evaluation of thyroid carcinoma.Methods : In September 2016 to September 2018 in Hunan Province People's Hospital and diagnosed with papillary thyroid carcinoma,selection of 43 patients with complete data,in which 3 samples were randomly selected for early preprocessing to obtain gene chip data,and R software was used to further analyze the gene chip data,from which differentially expressed genes were screened out.Then the protein interaction network analysis of differentially expressed genes was carried out in the online database,and the expression of target gene in papillary thyroid carcinoma and corresponding paracancerous tissues was obtained.Furthermore,the expression level of target gene was verified by immunohistochemistry in 40 cases of papillary thyroid carcinoma.Results:1.A total of 364 differentially expressed genes were screened by gene microarray in the 3 samples,including 282 differentially expressed genes that were down-regulated and 82 differentially expressed genes that were up-regulated,among which FN was the most up-regulated gene(Fold-change=94.12).A total of 228 differential LncRNAs were screened,among which 207 LncRNAs were down-regulated and 21 LncRNAs were up-regulated.2.Fifteen key genes,including ATP6V1 A,ATP6V1F,BCL2,BID,CFH,CFI,FN,ITGA8,ITGAV,ITGAV,ITGB1,KCNJ2,KCNQ1,SMAD9,SPARC and TGFBR1,were screened out,with FN as the center and significant correlation with ITGA8,ITGAV,ITGB1 and SMAD9.3.The interaction between FN gene and ECM-receptor in papillary thyroid carcinoma,focus adhesion,the role of proteoglycan in cancer,small cell lung cancer pathway,and extracellular matrix tissue,cell adhesion,extracellular corpuscle,Protein extracellular matrix,extracellular space,protease binding,heparin binding and other functions are related.4.The positive rate of FN was 85% in papillary thyroid carcinoma and42.5% in corresponding paracancerous tissues,the difference wasstatistically significant(P<0.05).5.The expression level of FN in papillary thyroid carcinoma was significantly higher than that in paracancerous tissue,the difference was statistically significant(P<0.05).Conclusions:1.The expression of FN gene in papillary thyroid carcinoma was significantly higher than that in the corresponding paracancerous tissues;2.FN gene may be related to the occurrence of papillary thyroid carcinoma.
