Expression And Significance Of KISS-1 And Matrix Metalloproteinase-9 In Ectopic And Eutopic Endometrium Of Endometriosis | Posted on:2007-12-26 | Degree:Master | Type:Thesis | Country:China | Candidate:Q W Shen | Full Text:PDF | GTID:2144360218953138 | Subject:Obstetrics and gynecology | Abstract/Summary: | PDF Full Text Request | Objective:To study the expressions of KISS-1 and matrix metalloproteinase-9 (MMP-9) in normal endometrium and in ectopic and eutopic endometrium in patients with endometriosis and to explore the relationship between KISS-1﹑MMP-9 and clinical phases.Methods:1. The expressions of KISS-1 and MMP-9 in 33 cases of normal endometrium as well as in 46 cases of ectopic endometrium and 16 cases of eutopic endometrium in endometriosis were detected by immunohistochemical EliVision plus method. Eutopic endometrium was obtained from 16 of these 46 endometriosis patients who were performed hysterectomy.2. On the basis of what we saw in the laparoscope or laparotomy ,we classified the 46 endometriosis patients into two groups (â… ï½žâ…¡s tage,Ⅲ~Ⅳstage), according to the classification of the revised American Fertility Society (r-AFS) in 1985, to compare the expressions of KISS-1 and MMP-9 between the two groups.3. The results were analyzed by Concise Statistics 2000 software.( including the positive expression rates, the positive intensity and the degree of staining of KISS-1 and MMP-9.)Results:1. The expressions of KISS-1 and MMP-9 in the tissues of normal endometrium and in ectopic /eutopic endometrium of endometriosis:There were positive expressions in the tissues of all the three groups, The positive expression rates of KISS-1 in ectopic and eutopic endometriotic tissues were significantly lower than those in the normal endometrium(P<0.01&P<0.05). In ectopic and eutopic endometrium of endometriosis and normal endometrium , the positive expression rates of KISS-1 were 30.43%(14/46), 31.25%(5/16)and 63.64%(21/33)respectively. The positive expression rates of MMP-9 in ectopic and eutopic endometriotic tissues were significantly higher than those in the normal endometrium(P<0.01&P<0.05). The positive rates of MMP-9 in the ectopic and eutopic endometrium of endometriosis and normal endometrium were 60.87%(28/46), 56.25%(9/16) and 27.27%(9/33)respectively. The positive intensity and the degree of staining of KISS-1 and MMP-9 in the three groups were compared too, the differences were significant.(P<0.01).2. The expression of KISS-1 and MMP-9 in different stages of endometriosis: The positive expression rates of KISS-1 in stageâ… ï½žâ…¡and in stageⅢ~Ⅳof endometriosis were 38.89%(7/18) and 25.00%(7/28) respectively; The positive expression rates of MMP-9 in the two groups were 44.44%(8/18) and 71.43%(20/28), respectively. There was no significant difference between the two groups.(P>0.05)Conclusions: 1. There were positive expressions of KISS-1 and MMP-9 in the tissues of normal endometrium and in ectopic /eutopic endometrium of endometriosis. The decreased expression of KISS-1 and the increased expression of MMP-9 protein may be involved in the invasion of endometriosis and they may be one of the reasons why ectopic and eutopic endometriatic cells have stronger invasive ability than that of normal endometric cells.2. The expressions of KISS-1 and MMP-9 in different stages of endometriosis were different, but there was no statistic value, indicatting that the expression level of KISS-1 and MMP-9 in tissues of ectopic endometrium may have nothing to do with different clinical phases. | Keywords/Search Tags: | Endometriosis, KISS-1, matrix metalloproteinases-9 (MMP-9), immunohistochemistry | PDF Full Text Request | Related items |
