Obejective:To study the therapeutic action and mechanism of Artesunate ontreating IgA Nephropathy model ratsMethods: Rat IgAN model was established by the method of"BSA+LPS+CCL4"; Rats were randomly assigned into five groups:nomal controlgroup, IgAN control group, small dose artesunate-treated group, large doseartesunate-treated group and ARB-treated group; The levels of 24-hour urineprotein, urine red blood cell, serum creatinine and transaminase were examined afterall the rats were killed; The renal tissue was detected by light microscopy andimmunofluorescence; The serum interleukin-2 and interleukin-6 were examined byELASIA; We detected the protein expression of TGF-β1 and MCP-1 byimmunohistochemistry; TGF-β1 and MCP-1 mRNA were examined with real-timePCR.Results:(1)The proteinuria and micro-hematuria were detected from the sixth weekand very obvious from eighth to tenth week; the proteinuria and hematuriadecreased after four weeks treatment, and these changes were more obvious in large dose artesunate-treated group and ARB-treated group; (2) The blood urea nitrogen(BUN)and serum creatinine(sCr)increased in IgAN control group, anddecreased in other groups; (3) The serum albumin(ALB) and total protein(TP)decreased obviously in in IgAN control group; the alanine aminotransferase(ALT)and aspartate aminotransferase(AST) were not changed in each group; (4)In renalbiopsy we can see diffuse mesangial cell proliferation, increased matrix and someglomeruli also show compensatory hypertrophy or sclerosis; lots of deposits of IgAwere detected in mesangial regions; above changes can increase after treatment; (5)The serum interleukin-2(IL-2) and interleukin-6(IL-6) increased in IgAN controlgroup, and decreased in each treamental group; (6)Compared with nomal controlgroup, the expression of TGF-β1 and MCP-1 by immunohistochemistry andreal-time PCR was higher in renal tissue of IgAN control group; and this changewas more obvious in large dose artesunate-treated group.Conlusion: Artesunate could lessen the proteinuria and hematuria of IgAnephropathy model rats, lessen the deposits of immunocomplex in renaltissue, diminish the hyperplasia of mesangial matrix; Artesunate could inhibit thelevel of serum interkeulin-2 and interkeulin-6, and inhibit the expression of TGF-β1and MCP-1 at the level of gene and protein in renal tissue.
