ACEI And Arb Joint Study On The Use Of Protective Effects On Kidney In Diabetic Rats

Objective: To detected the expressions of ED₁ antigen of rahagiocrine andmonocyte chemoatracttant protein-1(MCP-1) in glomerruli of diabetic rats byimmunohistochemical staining. To investigate the kidney-protective effects ofcombined use of Angiotesin-converting enzyme inhibitor(ACEI) andAngiotensinâ…¡ receptor antagonist(ARB) and its mechanism.Methods: The rats were randomly divided into five groups: normal controlrats(NC); streptozoein diabetic rats(DM); diabetic rats treated with small doseValsartan(V); diabetic rats treated with large dose Valsartan(V_L); diabetic ratstreated with Valsartan and Benazepril(VB). After 4,8 weeks ,blood glucose,blood urea nitrogen, creatinine clear rate, 24-hour urinary protein, kidneyweight/body weight ratio were measured. Expressions of ED₁ and MCP-1 ofglomeruli were detected by immunohistochemichal staining.Results: Blood urea nitrogen, creatinine clear rate, 24-hour urinary protein,kidney weight/body weight ratio, expressions of ED₁ and MCP-1 of DM ratswere significantly higher than that of NC rats., The above-mentioned indexsigns of DM rats of 8 weeks were higher than that of 4 weeks. 4 weeks, blood urea nitrogen, creatinine clear rate, 24-hour urinary protein,kidney weight/body weight ratio, expressions of ED₁ and MCP-1 of all treatedrats were significantly different with DM rats. 8 weeks, above-mentioned indexsigns of treated rats were lower than DM rats; That of V_L and VB were lowerthan V; 24-hour urinary protein, blood urea nitrogen and expressions of ED₁ ofVB were lower than VL. The relation between expressions of ED₁ and MCP-1 was positieelycorrelated with 24-hour urinary protein and creatinine clear rate.Conclusion: ARB has kidney-protective effectd on diabetic rats throughreducing expressions of ED₁ and MCP-1 of glomeruli. ACEI combined withARB can protect kidney of diabetic more strongly.
